超高效液相色谱-串联质谱法同时测定动物类中药材中6种伏马毒素

摘要 建立超高效液相色谱-串联质谱法同时测定动物类中药材中6种伏马毒素。样品经20 mL 80%乙腈水溶液提取,离心后上清液用QuEChERS EMR-Lipid净化包净化,净化液采用超高效液相色谱-串联质谱法在多反应监测模式下测定6种伏马毒素含量,色谱峰面积外标法定量。6种伏马毒素在质量浓度为2~40 ng/mL范围内线性关系良好,相关系数均大于0.999,检出限为0.4~1.4μg/kg。在16、40、80μg/kg 3个加标浓度水平下,平均回收率为77.3%~101.6%,相对标准偏差为1.7%~8.4%(n=6)。该方法简单快速,定性定量准确,可用于动物类中药材中伏马毒素的检测。 A method was established for the simultaneous determination of fumonisins in traditional Chinese animal medicines by ultra performance liquid chromatography-tandemmass spectrometry(UPLC-MS/MS).The samples were extracted with 20 mL 80%acetonitrile-water and purified by QuEChERS EMR-Lipid purification package after centrifugation.UPLC-MS/MS was employed for the quantification and confirmation of 6 fumonisins with the external standard method of chromatographic peak area.The linear relationships of 6 fumonisins was good in the range of 2-40 ng/mL,the correlation coefficients were all more than 0.999,the limit of detection were 0.4-1.4μg/kg.The average recoveries were 77.3%-101.6%with the relative standard deviations of 1.7%-8.4%(n=6)at three addition level of 16,40 and 80μg/kg.This method is simple and fast,accurate in qualitative and quantitative analysis,which can be used to detect fumonisins in traditional Chinese animal medicines.
