油莎豆粕小分子肽的制备、鉴定及功能分析 被引量:2

Preparation,identification and functional analysis of small molecular peptides from Cyperus esculentus meal

摘要 目的 研究油莎豆粕小分子肽的制备条件并对其组成与功能进行分析。方法 利用碱性蛋白酶与木瓜蛋白酶水解油莎豆粕蛋白质,同时通过液相色谱-质谱法分析多肽的序列,利用生物信息学技术分析多肽功能。结果 油莎豆粕小分子肽制备的最佳工艺参数为:酶解时间2.4 h、底物质量浓度2 mg/mL、加酶量4%、碱性蛋白酶:木瓜蛋白酶1:3,水解度为14.25%。对油莎豆粕小分子肽进行鉴定,共检测到30个肽段可信度高的油莎豆粕小分子肽分子,等电点范围是3.80~9.63,所有检测到的小分子肽分子均无毒性,具有调节血压、调节血糖、抗氧化的潜能。结论 经碱性蛋白酶与木瓜蛋白酶两种复合酶水解得到的油莎豆粕小分子肽具有潜在的调节血压、血糖、抗氧化功能,功能活性值均在0.6以上。本研究为油莎豆肽产品开发提供了理论依据和数据支持。 Objective To study the preparation conditions of small molecular peptides from Cyperus esculentus meal and analyze its composition and function.Methods The protein was hydrolyzed by alkaline protease and papain.The sequence of polypeptide was analyzed by liquid chromatography-mass spectrometry.The function of polypeptide was analyzed by bioinformatics.Results The optimum technological parameters for the preparation of small molecular peptides from Cyperus esculentus meal were as follows:The parameters included enzymatic hydrolysis time of 2.4 hours,substrate mass concentration of 2 mg/mL,enzyme dosage of 4%,alkaline protease:papain ratio of 1:3,resulting in a hydrolysis degree of 14.25%.The small peptides of Cyperus esculentus were identified.A total of 30 small molecule peptides of Cyperus esculentus meal were detected,exhibiting high credibility.The isoelectric point range of these peptides was found to be 3.80-9.63.Importantly,none of the detected small peptide molecules exhibited any toxic properties.It had the potential of blood pressure regulation,blood glucose regulation and anti-oxidation.Conclusion The small molecular peptides obtained by 2 kinds of complex enzymes alkaline protease and papain hydrolyzed have potential functions of regulating blood pressure,blood sugar and anti-oxidation.All functional activity values are above 0.6.This study provides the theoretical basis and data support for the development of peptides products.
