复方菊明对高血压伴高尿酸血症模型大鼠肾脏的影响

Effect of Compound Juming on Kidneys of Model Rats with Hypertension and Hyperuricemia

摘要 目的探讨复方菊明对高嘌呤饲料饲喂致高血压伴高尿酸血症模型大鼠尿酸水平的影响及肾脏保护作用。方法高血压模型大鼠50只,随机分为5组:模型对照组、复方菊明(10.0、5.0、2.5 g·kg^(-1))剂量组、阳性对照组(氯沙坦组);正常对照组Wistar大鼠,雄性,每组10只。采用连续饲喂高嘌呤饲料8周建立大鼠高血压伴高尿酸血症模型。每天给予高嘌呤饲料造模的同时给药,给药第3、6、8周眼眶取血测定血清尿酸水平,给药第2、5、7周测定4 h尿量及尿液中尿酸、肌酐、尿素氮、白蛋白、总蛋白、α1微球蛋白、β2微球蛋白的含量,并计算4 h尿酸排泄总量及尿酸清除率。结果复方菊明给药组能降低高血压伴高尿酸血症大鼠血清尿酸水平,升高尿液4 h尿量、尿酸、尿液肌酐、4 h尿酸排泄总量、尿酸清除率水平,降低尿液中白蛋白、α1微球蛋白、β2微球蛋白的水平。结论复方菊明具有显著的降尿酸作用及肾脏保护作用。 Objective To investigate the effects of compound Juming on urinary levels and renal function in a spontaneous hypertensive rat model with hyperuricemia induced by a high-purine diet.Methods Fifty spontaneous hypertensive rat were divided into the model control group,three compound Juming groups(10.0,5.0,2.5 g·kg^(-1)),and the positive control group(losartan group),with 10 rats in each group.The normal control group consisted of 10 Wistar rats.A spontaneous hypertensive rat model of hyperuricemia was established using continuous feeding of high-purine feed for 8 weeks.Compound Juming(10.0,5.0,2.5 g·kg^(-1))was administered intragastrically at the same time as modeling.Serum levels of uric acid were measured via orbital blood sampling in the 3rd,6th,and 8th weeks of administration.The 4-hour urine volume,levels of uric acid,creatinine,urea nitrogen,albumin,total protein,α1-microglobulin,andβ2-microglobulin in the urine were measured in the 2nd,5th,and 7th weeks of administration.Additionally,the 4-hour total rates of excretion and clearance of uric acid were calculated.Results The administration of compound Juming effectively reduced the level of serum uric acid in rats with hypertension and hyperuricemia,but increased the 4-hour urine volume,levels of uric acid and creatinine in the urine,4-hour total rates of excretion and clearance of uric acid,and decreased the levels of albumin,α1-microglobulin,andβ2-microglobulin in the urine.Conclusion Compound Juming has significant therapeutic effects against hyperuricemia,and is capable of renal protection.
