Ca^(2+)、Mn^(2+)离子对生防菌枯草芽孢杆菌CS16液体发酵过程产孢的影响

Effect on Spore Producing Ability of Biocontrol Bacterium Bacillus subtilis CS16:Calcium Ions and Manganese Ions in Liquid Fermentation Process

摘要 本文旨在通过单因素筛选和正交实验优化生防菌株CS16产芽孢发酵条件。利用单因素实验确定无机盐种类及添加量对CS16芽孢数和芽孢率的影响;通过正交实验进一步优化发酵工艺。结果表明,添加CaCl_(2)、MnSO_(4)·H_(2)O均可以提高CS16芽孢数,与对照相比达差异显著水平(P<0.05);正交实验结果显示在原有的发酵条件的基础上,同时添加0.8%CaCl_(2)和0.06%MnSO_(4)·H_(2)O培养48 h后,芽孢数最高达2.37×10^(9)cfu/mL,为单加CaCl_(2)时最高芽孢数的8.4倍,单加MnSO_(4)·H_(2)O最高芽孢数的6.5倍,与对照(CK)相比提高近20倍;同时可极大的提高芽孢率,达65.02%,相比对照提高近50%。Ca^(2+)、Mn^(2+)对CS16从营养细胞向芽孢状态转化具有明显的协同促进作用,同时添加Ca^(2+)、Mn^(2+)能有效的提高CS16液体发酵的芽孢数和芽孢率。 This study was conducted to optimize the fermentation conditions for spore production of strain Bacillus subtilis CS16 using the methods of single factor and orthogonal experiment.The effects of type and concentration of mineral salts on the spore count and sporulation rate of Bacillus subtilis CS16 were studied,and then fermentation technology was further optimized by orthogonal experiment on the basis of single-factor experiments.The results showed that the addition of CaCl_(2) or MnSO_(4)·H_(2)O in the medium significantly increased the spore count(P<0.05).Orthogonal experiment results indicated that the highest spore count reached 2.37×10^(9) cfu/mL after 48 h of fermentation when 0.8%of CaCl_(2) and 0.04%of MnSO_(4)·H_(2)O were simultaneously added,which was 8.4 times and 6.5 times of the highest spore count compared with CaCl_(2) and MnSO_(4)·H_(2)O were added alone,respectively.The spore count increased almost 20 times compared with the control group(CK),and the sporulation rate reached to 65.02%,which increased nearly by 50%compared with the CK group.Simultaneous addition of Ca^(2+)and Mn^(2+)had a synergistic effect on the spore formation of CS16,which could effectively increase the spore count and sporulation rate in CS16 liquid fermentation process.
