牡丹籽粕总黄酮提取工艺优化及降血脂功能研究 被引量:3

Optimization of extraction technology of total flavonoids from peony seed meal and its hypolipidemic function

摘要 采用单因素试验结合响应面法优化牡丹籽粕总黄酮提取工艺,并利用昆明小鼠高脂血症模型,对总黄酮降血脂活性进行了研究。试验结果表明:总黄酮提取最佳工艺条件为乙醇浓度50%、料液比1∶35、温度65℃、提取时间95 min,在此条件下测得牡丹籽粕总黄酮提取率为1.45%。与模型对照组相比,黄酮中、高剂量组小鼠血清中TC含量分别下降了19.6%(P<0.05)、31.6%(P<0.05),黄酮高剂量组小鼠血清中HDL-C含量上升了53.5%(P<0.01),而黄酮低、中、高剂量组小鼠血清中LDL-C含量分别降低了34.9%(P<0.05)、40.1%(P<0.01)、39.2%(P<0.01)。试验结果初步表明,该提取工艺稳定可行,总黄酮提取率高,且对小鼠脂质代谢紊乱症状具有一定的治疗作用。该研究可为牡丹籽粕黄酮降血脂活性的开发利用提供理论参考。 Single factor test combined with response surface method were used to optimize the extraction process of total flavonoids from peony seed meal,and the hypolipidemic activity of total flavonoids was studied by using Kunming mouse hyperlipidemia model.The results showed that the optimum extraction conditions were as follows:ethanol concentration 50%,ratio of material to liquid 1∶35,temperature 65℃,extraction time 95 min.Under these conditions,the extraction rate of totalflavonoids from peony seed meal was 1.45%.Compared with the model control group,the content of TC in serum of mice in middle and high dose offlavonoids decreased by 19.6%(P<0.05)and 31.6%(P<0.05),respectively,and the content of HDL-C in serum of mice in high dose offlavonoids increased by 53.5%(P<0.01).However,the content of LDL-C in serum of mice in low,middle and high dose groups decreased by 34.9%(P<0.05),40.1%(P<0.01)and 39.2%(P<0.01),respectively.The results preliminarily showed that the extraction process was stable and feasible,the extraction rate of totalflavonoids was high,and it had a certain therapeutic effect on the disorder of lipid metabolism in mice.This study can provide theoretical reference for the development and utilization of hypolipidemic activity offlavonoids from peony seed meal.
