摘要 体外核酸扩增技术在动物疫病病原检测中应用广泛。重组酶聚合酶扩增(RPA)作为一种新型的核酸等温扩增技术,可在较低温度(37~42℃)下扩增DNA或RNA,具有反应时间短、样品制备量少、特异性和敏感性高等优点。RPA突破了常规聚合酶链反应(PCR)实施时对实验室条件的依赖性,并能高效和灵敏地检测动物疫病病原。基于RPA技术的病原菌检测方法可实现对动物细菌病的现场快速诊断。论文综述了RPA技术的反应体系和条件、反应原理和产物检测方法等,介绍了RPA技术在动物病原菌检测中的应用和潜在价值,以期为动物疾病快速诊断研究提供参考。 Technology of nucleic acid amplification in vitro is widely used in animal epidemic disease detection.As a new isothermal amplification technique,RPA can amplify DNA or RNA at a constant low temperature(37-42℃),with the advantages of short reaction time,minimal sample preparation,high specificity and sensitivity.Recombinase polymerase amplification technology(RPA)breaks through the dependence of traditional polymerase chain reaction(PCR)on laboratory conditions,and it is becoming a method for the effective and sensitive detection of pathogens of animal epidemics.The pathogen detection method based on RPA technology can achieve rapid on-site diagnosis of animal bacterial diseases.In this paper,the composition of reaction system,the reaction conditions,the reaction principle,and the detection methods of amplified products of RPA were introduced,and the application and potential value of RPA in the detection of animal bacterial diseases were also introduced,aiming at providing the reference for the rapid diagnosis of animal diseases.
