摘要
为了对临床上马立克病(MD)准确诊断,研究利用杆状病毒表达系统表达马立克病病毒(MDV)强毒株Meq重组蛋白免疫小鼠,利用杂交瘤技术研制MDV Meq蛋白特异性单克隆抗体,并以此为基础建立MD诊断方法。结果显示:试验成功制备了4株Meq蛋白特异性单克隆抗体,分别命名为MDV-Meq-1D6、MDV-Meq-1D10、MDV-Meq-6B3、MDV-Meq-6D10;Western blot结果证明这些单克隆抗体不仅能与体外表达的Meq蛋白反应,而且能识别MD肿瘤细胞系MSB-1中的Meq蛋白和MD肿瘤组织细胞中的Meq蛋白;以Meq蛋白单克隆抗体成功建立免疫组化诊断MD的方法,该方法特异性强,结果稳定,易观察判定。研究表明,试验制备了4株Meq蛋白单克隆抗体(MDV-Meq-1D6、MDV-Meq-1D10、MDV-Meq-6B3、MDV-Meq-6D10),基于此建立的免疫组化MD诊断方法为临床MD鉴别诊断提供技术支撑。
In order to accurately clinical diagnose Marek's disease(MD),mice were immunized with Marek's disease virus(MDV)virulent strain Meq recombinant protein expressed by baculovirus expression system,and monoclonal antibodies specific to MDV Meq protein were prepared by hybridoma technology.With the monoclonal antibody,a diagnosis method for MD was es⁃tablished.The results showed that four monoclonal antibodies specific to Meq protein were successfully developed,which were named MDV-Meq-1D6,MDV-Meq-1D10,MDV-Meq-6B3 and MDV-Meq-6D10,respectively.These monoclonal antibodies could not only react with Meq protein expressed in vitro,but also recognize Meq protein in MD tumor cell line MSB-1 and Meq protein in MD tumor tissue.With the monoclonal antibody,an immunohistochemical method for MD diagnosis was established,which showed strong specificity,stable results,and was easy to observe and determine.It's suggested that four monoclonal antibodies(MDV-Meq-1D6,MDV-Meq-1D10,MDV-Meq-6B3 and MDV-Meq-6D10)specific to Meq protein were prepared and the es⁃tablished immunohistochemical method for MD could provide technical support for differentially clinical diagnosis of MD.
