蒲公英总黄酮的提取工艺优化及抗氧化活性评价

摘要 本文采用乙醇浸提法从蒲公英中提取总黄酮,考察了乙醇浓度、提取温度、提取时间等因素对蒲公英总黄酮提取效果的影响。以总黄酮含量为指标,在单因素实验基础上采用正交实验法对提取工艺进行优化。最佳工艺条件为:乙醇浓度为40%,提取温度为70℃,提取时间为80min,此条件下蒲公英提取物总黄酮含量为0.9170mg·mL^(-1)。通过DPPH清除实验、羟基自由基清除实验,对蒲公英总黄酮提取物进行抗氧化活性评价,其中DPPH自由基清除率IC_(50)值为0.8492mg·mL^(-1),羟基自由基的清除能力IC_(50)值为1.0717mg·mL^(-1)。该结果表明,蒲公英总黄酮提取物具有良好的抗氧化清除自由基能力。 Total flavonoids were extracted from dandelion by ethanol extraction.The effects of ethanol concentration,extraction temperature,extraction time and other factors on the extraction efficiency of total flavonoids were investigated.The extraction process was optimized by orthogonal experiment based on single factor experiment.The optimal conditions were ethanol concentration of 40%,extraction temperature of 70℃,extraction time of 80min,under these conditions,total flavonoids content of dandelion extract was 0.9170mg·mL^(-1).The antioxidant activity of total flavonoids from dandelion was evaluated by DPPH scavenging experiment and hydroxyl free radical scavenging experiment.The IC_(50) value of DPPH free radical scavenging was 0.8492mg·mL^(-1),and the IC_(50) value of hydroxyl free radical scavenging ability was 1.0717mg·mL^(-1).The results showed that the flavonoid extract of dandelion had good antioxidant ability in scavenging free radicals.
