NtCycB2和NtJAZ1共敲除对烟草腺毛密度和烟碱含量的影响

摘要 为了提高烟草的腺毛密度、腺毛分泌物和烟碱含量,从而改善烟株抗性和烟叶品质,以NtCycB2突变的多腺毛K326突变株系(hairy K326)为材料,采用CRISPR/Cas9技术对NtJAZ1进行基因编辑,筛选出NtCycB2与NtJAZ1基因共敲除的纯合株系J-21,并以K326和hairy K326为对照,对J-21的农艺性状、腺毛发育、烟碱生物合成和烟叶化学成分进行了研究。结果表明,J-21植株生长发育正常,主要农艺性状与K326和hairy K326无显著差异,其叶片腺毛密度比hairy K326有所降低,但比K326显著提高,腺毛分泌物含量(质量分数)提高156.35%。J-21叶片烟碱含量在现蕾期比K326和hairy K326显著提高,在烟株打顶后提高幅度明显降低,成熟期叶片烟碱含量比K326和hairy K326分别提高37.14%和32.28%。J-21烤后烟叶化学成分协调,烟碱含量比K326和hairy K326分别提高39.81%和45.32%,西柏烷类降解产物茄酮含量比K326和hairy K326分别提高217.49%和48.45%。表明NtCycB2和NtJAZ1共敲除在不影响烟株生长发育的前提下,可有效提高烟草叶片腺毛密度、促进腺毛分泌物和烟碱的积累,是进行烟草抗性和品质聚合改良的有效途径之一。 In order to increase the density of glandular trichomes and nicotine content of flue-cured tobacco to improve plant resistance and tobacco quality,hairy mutant line K326 with NtCycB2 knockout was used as the study material,and NtJAZ1 gene was edited by CRISPR/Cas9 technology.NtCycB2 and NtJAZ1 gene co-knockout line J-21 was selected and its agronomic traits,glandular trichome development,nicotine biosynthesis and leaf chemical components were studied with K326 and hairy K326 as controls.The results showed that the growth and development of J-21 plants were normal,and its main agronomic characteristics were not significantly different from those of K326 and hairy K326.The density of leaf glandular trichomes of J-21 was lower than that of hairy K326,but significantly higher than that of K326,and the content(mass fraction)of glandular trichome secretion was increased by 156.35%.At the bud stage,the nicotine content of J-21 was significantly higher than that of K326 and hairy K326,but the rate of increase decreased after topping.The nicotine content in the mature stage was 37.14%and 32.28%higher than those of K326 and hairy K326,respectively.The chemical components in cured leaves were harmonious,and the nicotine content of J21 was increased by 39.81%and 45.32%,the content of cembranoid degradation product solanone was increased by 217.49%and 48.45%compared with K326 and hairy K326,respectively.These results indicated that the co-knockout of NtCycB2 and NtJAZ1 can effectively increase the glandular trichome density of tobacco leaves,promote the accumulation of glandular trichome secretion content and nicotine without affecting the normal growth and development of tobacco plants,which is one of the effective methods to simultaneously improve tobacco resistance and characteristics.
