清血解毒合剂对小鼠变应性接触性皮炎的效果评价

Efficacy evaluation of Qingxue Jiedu Mixture on allergic contact dermatitis in mice

摘要 目的:探索清血解毒合剂对变应性接触性皮炎(allergic contact dermatitis,ACD)小鼠的干预作用并预测其药效作用的关键成分。方法:采用方酸二丁酯诱导小鼠构建ACD模型,将小鼠随机分为空白组,模型组,阳性组,清血解毒合剂低、中、高剂量组。在治疗过程中,记录小鼠抓挠和颈部皮肤状况。通过HE和甲苯胺蓝染色切片观察皮肤组织病理学变化,ELIA法检测血清TNF-α、IL-1β水平,RT-qPCR法检测皮肤组织TNF-α、IL-1β、IFN-γ、IL-4 mRNA表达水平,Western blot法检测皮肤组织p-ERK、p-P38蛋白表达。通过数据库预测清血解毒合剂的关键成分和ACD核心靶点,利用分子对接验证。结果:清血解毒合剂能减少小鼠抓挠次数,减轻颈部皮肤红斑、水肿、鳞屑覆盖,降低皮层厚度和肥大细胞浸润,降低血清TNF-α、IL-1β水平和皮肤组织TNF-α、IL-1β、IFN-γ、IL-4 mRNA的表达量,抑制皮肤组织p-ERK、p-P38蛋白表达水平。分子对接结果显示槲皮素、β-谷甾醇与核心靶点具有较强结合力。结论:清血解毒合剂中槲皮素、β-谷甾醇、黄芩素等关键成分能协同改善ACD小鼠的症状,其作用可能与抑制MAPK信号通路有关。 OBJECTIVE To explore the intervention of Qingxue Jiedu Mixture in allergic contact dermatitis(ACD) mice,and to predict the key components for its pharmacodynamic effect.METHODS Mice were induced with dibutyl acetate to construct an ACD model,and randomly divided into control group,model group,positive group,and Qingxue Jiedu Mixture low-dose,medium-dose and high-dose groups.During the treatment,the scratching and neck skin condition of mice were recorded.The histopathological changes of skin were observed by HE and toluidine blue stained sections,the serum TNF-α and IL-1β levels were detected by ELISA,the TNF-α,IL-1β,IFN-γ and IL-4 mRNA expression levels in skin tissue were detected by RT-qPCR,and the p-ERK and p-P38 protein expression in skin tissue were detected by Western blot.The database was used to predict the key components of Qingxue Jiedu Mixture and the core targets of ACD,and the results were validated by molecular docking.RESULTS Qingxue Jiedu Mixture could reduce the number of scratching and neck skin erythema,edema and scale coverage,decrease the thickness of the skin layer and mast cell infiltration,reduce the levels of serum TNF-α and IL-1β,and the expression of TNF-α,IL-1β,IFN-γ and IL-4 mRNA in skin tissues,and inhibit the expression levels of p-ERK and p-P38 protein in skin tissue.The molecular docking results showed strong binding between quercetin and beta-sitosterol and the core targets.CONCLUSION Qingxue Jiedu Mixture synergistically improves the symptoms in ACD mice through the key components such as quercetin,beta-sitosterol and baicalein,and its action may be related to the inhibition of MAPK signaling pathway.
