基于PI3K/Akt通路研究蛇伤胶囊调节血小板活化功能治疗竹叶青蛇伤凝血障碍机制

摘要 目的基于PI3K/Akt通路研究蛇伤胶囊调节血小板活化功能治疗竹叶青蛇伤凝血障碍机制。方法24只新西兰大白兔被随机分为正常对照组、模型组、蛇伤胶囊组,每组8只雌雄各半,根据前期研究方法模型组和蛇伤胶囊组以兔耳缘静脉注射0.75 mg/kg竹叶青蛇毒液,建立竹叶青蛇伤兔模型,正常对照组注射等量生理盐水。注射后6 h后予正常对照组和模型组以10 ml/(kg·d)生理盐水灌胃,蛇伤胶囊组予10 ml/(kg·d)蛇伤胶囊所配药液灌胃。以上3组连续灌胃1周后兔耳缘静脉采血5 ml,血小板分离提取后Western blot检测各组血小板细胞PTEN、PI3K、Akt蛋白表达,qPCR检测各组血小板活化指标CD62P mRNA表达。结果与正常对照组相比,模型组血小板细胞PTEN蛋白表达增加(P<0.01),PI3K、Akt蛋白及CD62P mRNA表达减少(P<0.01);与模型组相比,蛇伤胶囊组血小板细胞PTEN蛋白表达减少(P<0.01),PI3K、Akt蛋白及CD62P mRNA表达增加(P<0.01)。结论蛇伤胶囊可通过上调PI3K/Akt通路,改善竹叶青蛇伤导致的血小板活化功能受抑制,从而治疗竹叶青蛇伤凝血障碍。 Objective Based on the PI3K/Akt pathway,to study the mechanism of Sheshang capsule regulating platelet activation function in the treatment of coagulation disorders caused by bamboo leaf green snake injury.Methods Twenty-four New Zealand white rabbits were randomly divided into a normal control group,a model group,and a snake wound capsule group,Each group consisted of eight males and half females.According to the previous research method,the model group and the snake wound capsule group were injected with 0.75 mg/kg bamboo leaf green snake venom into the rabbit ear vein to establish a bamboo leaf green snake wound rabbit model,while the normal control group was injected with an equal amount of physiological saline.6 h after injection,normal control group and model group were given 10 ml/(kg·d)normal saline intragastric administration,and Shesheng capsule group was given 10 ml/(kg·d)Shesheng capsule intragastric administration.After continuous intragastric administration for 1 week,5 mL blood was collected from the auricular vein of rabbits in the above 3 groups.After platelet separation and extraction,Western blot was used to detect the expression of PTEN,PI3K,and Akt proteins in platelet cells in each group.qPCR was used to detect the expression of CD62P mRNA,a platelet activation indicator in each group.Results Compared with the normal control group,the expression of PTEN protein in platelet cells of the model group was increased(P<0.01),while the expression of PI3K,Akt protein,and CD62P mRNA was reduced(P<0.01);Compared with the model group,the expression of PTEN protein in platelet cells in the Sheshang capsule group was reduced(P<0.01),while the expression of PI3K,Akt protein,and CD62P mRNA was increased(P<0.01).Conclusions Snake wound capsule can improve the inhibition of platelet activation function caused by bamboo leaf green snake injury by upregulating the PI3K/Akt pathway,thereby treating coagulation disorders caused by bamboo leaf green snake injury.
