摘要
为了研究诃子、金银花、黄连等26种单味中药对鲤(Cyprinus carpio)源嗜水气单胞菌(Aeromonas hydrophila)体外抑菌活性,该试验采用水煮方法提取诃子、金银花、黄连等26种单味中草药的有效成分,调整中草药药液浓缩为1g/mL(每1mL药液含1g生药),以临床分离鲤的源致病性嗜水气单胞菌(HT333株)为试验菌株,利用平板打孔法和微量二倍稀释法分别测定诃子、金银花、黄连等26种单味中草药对鱼源致病性嗜水气单胞菌(HT333株)的抑菌圈直径和最小杀菌浓度(MBC)。结果显示,对诃子、五味子、金银花等3种极度敏感,对黄连、苦参、板蓝根、黄芩等4种药物高度敏感,对苏木、五倍子、青皮等9种药物中度敏感,对地丁、白头翁、穿心莲3种药物低度敏感。对诃子、五味子、金银花、黄连、苦参、板蓝根、黄芩等7种药物的MBC在7.825mg/mL~15.65mg/mL之间,对沙棘、五倍子、秦皮3种药物的MBC在31.25mg/mL~62.5mg/mL之间,对青皮、马齿苋、黄柏等8种药物的MBC在125mg/mL~250mg/mL之间,对其他药物则无效果。该研究筛选出了诃子、五味子、金银花、黄连、苦参、板蓝根、黄芩等7种对鲤的源致病性嗜水气单胞菌敏感的中药,为进一步进行组方及中草药防控鲤的嗜水气单胞菌病提供研究基础。
In order to study the in vitro bacteriostatic activity of 26 single herbs such as chebula,Lonicera japonica,Coptis chinensis against Cyprinus carpio source Aeromonas hydrophila,the assay was conducted on chebula,Lonicera japonica,Coptis chinensis and 26 other single herbs,using the boiling method to extract the active ingredients,adjusting the herbal medicinal liquid concentrated to 1g/mL(each 1mL medicinal liquid contains 1g of raw herbs),the clinical isolation of carpio source pathogenic Aeromonas hydrophila(HT333 strain)as the test strains,the use of plate perforation method and micro two-fold dilution method to determine respectively.The diameter of the circle of inhibition and the minimum bactericidal concentration(MBC)of 26 single herbs,including chebula,Lonicera japonica,and Coptis chinensis,against the fish-derived pathogenic Aeromonas hydrophila(strain HT333)were determined using the plate punch method and the microdouble dilution method,respectively.The results showed extreme sensitivity to three species of chebula,Schisandra chinensis and Lonicera japonica,and extreme sensitivity to four species of Coptis chinensis,Sophora flavescens,Isatis tinctoria and Scutellaria baicalensis Georgi and 4 other drugs are highly sensitive,moderately sensitive to 9 drugs such as Caesalpinia sappan Linn.,Rhus chinensis Mill.and Citrus reticulata Blanco,and Corydalis bungeana,Pulsatilla chinensis,Andrographis paniculata and 3 other drugs with low sensitivity.To seven drugs such as chebula,Schisandra chinensis,Lonicera japonica,Coptis chinensis,Sophora flavescens,Isatis tinctoria,Scutellaria baicalensis Georgi and others the MBC ranged from 7.825mg/mL to 15.65mg/mL,to Hippophae rhamnoides Linn.,Rhus chinensis,and Cortex fraxini 3 drugs ranged from 31.25mg/mL to 62.5mg/mL,and to Citrus reticulata Blanco,Portulaca oleracea,Phellodendron chinense Schneid and 8 other drugs with MBC ranging from 125mg/mL to 250mg/mL,while it was ineffective against other drugs.This study screened chebula,Schisandra chinensis,Lonicera japonica,Coptis chinensis,Sophora flavescens,Isatis tinctoria,Scutellaria baicalensis Georgi Seven Chinese herbal medicines sensitive to Aeromonas hydrophila,the source pathogen of common carp,were used to provide a research basis for further formulas and herbal remedies for the prevention and control of Aeromonas hydrophila disease in common Cyprinus carpio.
