摘要
目的:探讨夹脊电针对脊髓损伤大鼠铁死亡的影响,明确电针促进脊髓损伤修复的作用机制。方法:72只大鼠随机分为假手术组(Sham)、模型组(SCI)、夹脊电针组(EA)和抑制剂组(Fer-1)4组,每组18只,再将每组按照不同的治疗时间分为3 d组、7 d组与14 d组3个亚组,每亚组各6只。分别治疗3 d、7 d与14 d进行BBB评分、斜板实验,之后对损伤节段脊髓组织进行HE染色、尼氏染色、Western Blot和Elisa检测,观察铁死亡通路相关因子GSH、GPX4的表达情况及神经元数量。结果:与Sham组比较,SCI组3 d、7 d与14 d的BBB评分与斜板角度降低,差异有统计学意义(P<0.01),脊髓前角神经元显著减少,差异有统计学意义(P<0.01),GSH、GPX4表达显著降低,差异有统计学意义(P<0.01);与SCI组比较,EA组3 d、7 d与14 d的BBB评分与斜板角度显著增加,差异有统计学意义(P<0.05),脊髓前角神经元显著增加,差异有统计学意义(P<0.01),GSH表达显著增加,差异有统计学意义(P<0.01),GPX4表达在7 d、14 d显著增加,差异有统计学意义(P<0.01);与SCI组比较,Fer-1组3 d、7 d与14 d的BBB评分与斜板角度显著增加,差异有统计学意义(P<0.05),脊髓前角神经元显著增加,差异有统计学意义(P<0.01),GSH、GPX4表达显著增加,差异有统计学意义(P<0.01);与Fer-1组比较,EA组BBB评分与斜板角度在14 d显著增加,差异有统计学意义(P<0.05),脊髓前角神经元无明显改变,差异无统计学意义(P>0.05),GSH、GPX4表达无明显变化,差异无统计学意义(P>0.05)。结论:脊髓损伤后铁死亡加剧,电针治疗能够有效抑制Xc^(-)/GPX4通路促进脊髓损伤大鼠后肢运动功能恢复。
Objective:To investigate the effect of Jiaji electro-acupuncture(EA)on ferroptosis in rats with spinal cord injury(SCI)and to clarify the action mechanism of EA in promoting SCI repair.Methods:72 rats were randomly divided into the sham operation group,the model group,the Jiaji EA group and the inhibitor(Fer-1)group,with 18 rats in each group;each group was then divided into three subgroups according to different treatment time,including 3 d group,7 d group and 14 d group,with 6 rats in each subgroup.BBB scores and slant plate experiments were performed on 3 d,7 d and 14 d of treatment,followed by HE staining and Nissl's staining,and Western Blot and Elisa assays on spinal cord tissues at the injury stage were applied to observing the expressions of ferroptosis pathway-related factors GSH and GPX4 and the number of neurons.Results:Compared with those in the sham operation group,the BBB scores and the oblique plate angles decreased significantly on 3 d,7 d and 14 d(P<0.01),and the neurons in the anterior horn of the spinal cord decreased significantly(P<0.01),and the expressions of GSH and GPX4 decreased significantly in the model group(P<0.01).Compared with those in the model group,the BBB scores and the oblique plate angles increased significantly on 3 d,7 d and 14 d(P<0.05),the neurons in the anterior horn of the spinal cord increased significantly(P<0.01),the expression of GSH increased significantly(P<0.01),and the expression of GPX4 increased significantly on 7 d and 14 d in the EA group(P<0.01).Compared with those in the model group,the BBB scores and the oblique plate angles significantly increased on 3 d,7 d and 14 d(P<0.05),the neurons in the anterior horn of the spinal cord significantly increased(P<0.01),and the expressions of GSH and GPX4 significantly increased in the inhibitor group(P<0.01).Compared with those in the inhibitor group,the BBB scores and the oblique plate angles significantly increased on 14 d in the EA group(P<0.05),but there were no statistical differences in the neurons in the anterior horn of the spinal cord(P>0.05),also there were no statistical differences in the expressions of GSH and GPX4 between the EA group and the inhibitor group(P>0.05).Conclusion:Ferroptosis is increased after SCI,and EA treatment can effectively inhibit the Xc^(-)/GPX4 pathway to promote the recovery of motor function of the hind limbs in the rats with SCI.
