超氧化物歧化酶1基因与果蝇寿命和攀爬能力的关系

摘要 目的观察超氧化物歧化酶1(superoxide dismutase 1,SOD1)基因(sod1)表达对果蝇寿命和运动能力的影响,分析sod1与衰老之间的联系。方法利用RNAi干扰技术,获得sod1敲低果蝇;运用载体构建和显微注射的技术获得sod1过表达效应;通过免疫组织化学的手段定位sod1表达产物。结果sod1表达产物在细胞质中高表达,在细胞核中不表达;sod1全身性敲低果蝇与正常组果蝇相比表现出显著的寿命缩短以及明显的攀爬能力下降现象,而sod1过表达结果则恰恰相反。结论SOD1调控果蝇的衰老和运动能力。 Objective To observe the effects of superoxide dismutase 1(SOD1)gene(sod1)expression on the lifespan and motor abilities of Drosophila and to analyze the relationship between sod1 and aging.Methods RNA interference(RNAi)technology was utilized to achieve sod1 knockdown in Drosophila;vector construction and microinjection techniques were used to induce sod1 overexpression;and immunohistochemistry was employed to localize sod1 expression product.Results The expression product of sod1 was highly expressed in the cytoplasm but not in the nucleus.Compared to the normal group,Drosophila with systemic sod1 knockdown showed a significant reduction in lifespan and a noticeable decrease in climbing ability.In contrast,the results were opposite in the case of sod1 overexpression.Conclusion SOD1 plays a crucial role in regulating the aging and motor abilities of Drosophila.
