4株不同基因型新城疫病毒毒力测定及与商品疫苗株抗原性比较

Virulence Determination of Four Newcastle Disease Viruses of Different Genotypes and Comparison of Their Antigenicities with Commercial Vaccine Strains

摘要 为了探究不同基因型和不同来源4株新城疫病毒(Newcastle disease virus,NDV)及现有商品疫苗株的抗原性差异,测定4株新城疫病毒的鸡胚最小致死量的平均死亡时间(mean death time,MDT)和1日龄雏鸡脑内接种致病指数(intracerebral pathogenicity index,ICPI),分别制备油乳苗,再与商品疫苗NDV La Sota株、A-Ⅶ株分别接种SPF鸡,收集单因子血清进行HI交叉试验和细胞交叉中和试验。结果显示,基因Ⅵ型NDV B27为中等毒力毒株,基因Ⅶ型NDV B9、基因Ⅸ型NDV B14、基因Ⅻ型NDV GX01为强毒株;HI交叉试验结果显示,基因Ⅵ型NDV B27和基因Ⅸ型NDV B14抗原同源性为0.64,抗原性差异微小,其他毒株之间的抗原同源性为0.69~0.99,抗原差异不大;细胞交叉中和试验结果显示,基因Ⅵ型NDV B27和基因Ⅸ型NDV B14抗原同源性为0.42,基因Ⅵ型NDV B27和基因Ⅱ型疫苗株La Sota抗原同源性为0.47,抗原差异较大,其余毒株及疫苗株La Sota之间抗原差异微小。试验结果可为后续筛选NDV疫苗候选株提供参考。 The aim is explore the antigenic differences between four Newcastle disease viruses(NDV)of different genotypes and sources with the commom commercial vaccine strains.Four NDVs of different genotypes were prepared for vaccines with oil emulsion after evaluating the pathogenicity indexes of MDT and ICPI,and SPF chickens were immunized with these four vaccines and commercial vaccines La Sota and A-Ⅶ,respectively.Single-factor serum was collected for HI cross test and cell cross neutralization test.The virulence index showed that genotypeⅥNDV B27 was moderately virulent,while genotypeⅦNDV B9,genotypeⅨNDV B14 and genotypeⅫNDV GX01 were virulent.The HI cross tests showed that the antigenic homology between genotypeⅥNDV B27 and genotypeⅨNDV B14 was 0.64,and the antigenic difference between the other NDV was between 0.69 and 0.99,and there was no significant antigenicity difference.The antigenic homology between genotypeⅥNDV B27 and genotypeⅨNDV B14 was 0.42.The antigenic homology between genotypeⅥNDV B27 and genotypeⅡvaccine strain La Sota was 0.47.There were few antigenic differences between the remaining NDVs and La Sota strain.The results of this study provide a research basis for screening candidate NDV vaccine strain.
