摘要
为研究和开发蜜本南瓜EST-SSR标记,本试验以蜜本南瓜为材料,经RNA提取、cDNA文库构建和转录组测序,获得36.96 Gb序列数据,组装并去冗余后得到157 943条unigene。使用MISA (v1.0)软件进行SSR检测,结果表明59 365个SSR位点分布于41 097条unigene中,SSR分布频率为37.59%,发生频率为26.02%,平均分布距离为4 672 bp。单核苷酸、二核苷酸和三核苷酸重复所占比例最高,三者占EST-SSR总数的90.37%,其中以二核苷酸重复最多(30.62%),其优势重复基序为AG/CT。以MB21、2202、M10B三个南瓜材料的DNA为模板进行多态性分析,结果表明30对引物中6对引物的扩增产物表现出良好的多态性。本研究开发了蜜本南瓜EST-SSR标记并分析其多态性,可为南瓜品种分子鉴定、遗传图谱构建和遗传多样性分析等提供理论基础。
In order to research and develop EST-SSR markers for Miben pumpkin,36.96 Gb data were obtained after RNA extraction,cDNA library construction and transcriptome sequencing,and 157943 unigenes were de-tected after assemblingand redundancyremoval in this experiment.SSR detection was performed using MISA(v1.0)software,and the results showed that 59365 SSR loci were distributed in 41097 unigenes,the SSR distribution frequency was 37.59%,its occurrence frequency was 26.02%,and its average distribution distance was 4672 bp.Single nucleotide,dinucleotide and trinucleotide repeats accounted for high proportion,accounting for 90.37%of the total EST-SSR,of which dinucleotide repeats were the most(30.62%),and the dominant repeat motif was AG/CT.The DNA of three pumpkin materials MB21,2202 and M10B were used as templates for polymorphism analy-sis.The amplified products of 6 among 30 pairs of primers showed polymorphism.The development and polymor-phism analysis of EST-SSR markers in Miben pumpkin would lay foundation for molecular identification,genetic map construction and genetic diversity analysis of pumpkin varieties.
