塔本文都苏提取物对糖尿病肾病小鼠肾功能的改善和保护作用

Improvement and Protection Effects of TBWDS Extract on Renal Function in Diabetic Nephropathy Mice

摘要 为观察蒙药方剂塔本文都苏(TBWDS)提取物对糖尿病肾病小鼠肾功能的保护作用,小鼠腹腔注射链脲佐菌素联合高脂高糖饲料喂养构建糖尿病肾病(DN)模型。将模型小鼠随机分为模型组、TBWDS高剂量组、TBWDS低剂量组、二甲双胍组、正常对照组,每组10只。TBWDS高、低剂量组分别灌胃8 g/kg和4 g/kg TBWDS提取物,二甲双胍组灌胃0.3 g/kg二甲双胍,模型组和正常对照组灌胃等量生理盐水。治疗12周后,进行血、尿和肾组织生化指标检测以及肾脏组织石蜡切片的HE染色、六胺银染色和TGF-β1免疫组化染色。与模型组比较,高、低剂量的TBWDS提取物均可使血清UA、BUN、Scr及尿mAlb水平降低,使肾组织中的ROS和MDA含量降低,而GSH-Px和SOD的活性升高(P<0.01),同时使肾组织的病理损伤明显改善,TGF-β1的表达减弱,高剂量TBWDS提取物的作用尤为明显,与二甲双胍的作用无明显差异。表明TBWDS提取物能够有效缓解DN小鼠的肾损伤,从而改善肾功能。TBWDS有望开发成为针对DN患者的治疗方剂。 To observe the protective effect of Tabenwendusu(TBWDS)extract on renal function in diabetic ne⁃phropathy mice,the diabetic nephropathy(DN)model was established by intraperitoneal injection of streptozotocin and feeding with high fat and high sugar diet.The model mice were randomly divided into model group,TBWDS high-dose and low-dose groups,metformin group and normal control group,10 in each group.The high-dose and low-dose TBWDS groups were given 8 g/kg and 4 g/kg TBWDS extract respectively,the metformin group was given 0.3 g/kg metformin,and the model group and the normal control group were gavaged with the same amount of nor⁃mal saline.After 12 weeks of treatment,the biochemical indexes of blood,urine and renal tissue were detected,as well as HE staining,hexamine silver staining and transforming growth factorβ1(TGF-β1)immunohistochemical staining of paraffin-embedded renal sections.Compared with the model group,both high-dose and low-dose of TBWDS extract could reduce the levels of blood UA,BUN,Scr,and urine mAlb,reduce the contents of ROS and MDA in kidney tissue,and increase the contents of GSH-Px and SOD(P<0.01);at the same time,significantly im⁃prove the pathological damage of renal tissue and decrease the expression of TGF-β1,the effect of high-dose TBWDS extract was particularly obvious,and was not significantly different from that of metformin.The results indi⁃cated the TBWDS extract could effectively alleviate the renal injury in DN mice and improve renal function.TBWDS is expected to be developed into a therapeutic prescription for patients with DN.
