摘要
【目的】揭示毛竹Phyllostachys edulis种子发育过程中形态和生理特性的动态变化规律,为人工进行宏观调控、获得优质毛竹种子提供理论参考。【方法】以桂林海洋山一带的开花毛竹为材料,采集开花后不同时期的毛竹种子,对其发育过程中的形态和生理指标进行观察测定。【结果】1)花后10~30 d毛竹种子胚和胚乳细胞分裂分化活动剧烈,种子长、宽、厚等性状增长速度较快;花后40~60 d种子胚和胚乳细胞形态发育成熟,种子长、宽、厚等性状增长速度减缓。2)不同发育时期毛竹种子的可溶性糖、粗淀粉及可溶性蛋白含量存在显著性差异(P <0.05),可溶性糖与粗淀粉含量均呈单峰曲线变化,可溶性蛋白含量在4.89%~5.82%范围内波动变化。3)不同发育时期毛竹种子的活性氧含量和抗氧化酶活性具有显著性差异(P <0.05),H_(2)O_(2)含量与POD、CAT活性呈单峰曲线变化,而O_(2)^(-)含量与SOD活性变化趋势相似。4)不同发育时期毛竹种子的内源激素含量存在显著性差异(P <0.05),IAA、GA_(3)及ZR含量均呈先升后降再升的趋势,ABA含量则呈单峰曲线变化,(IAA+GA_(3)+ZR)/ABA的比值呈先降后升的趋势,在花后40 d达到最小值,为1.001。【结论】花后10~30 d抗氧化酶活性以及内源激素含量逐渐升高,有效促进毛竹种子细胞代谢与分裂分化;花后30~60 d毛竹种子的分化活动基本结束并进入营养积累阶段,该时期抗氧化酶活性和内源激素逐渐下降,营养物质含量进一步增加。
【Objective】The dynamic changes of morphological and physiological characteristics of Phyllostachys edulis seeds during their development were revealed,which provided reference for macro-control and obtaining high-quality seeds.【Method】The seeds of P.edulis were collected from Yangshan area of Guilin.The morphological and physiological indexes of P.edulis were observed and measured in different periods after flowering.【Result】(1)From 10 to 30 days after flowering,the embryo and endosperm cells of P.edulis seeds had vigorous activity of division and differentiation,and the length,width and thickness of seeds increased rapidly.The seed embryo and endosperm cells entered the mature stage in 40 to 60 days after flowering,and the growth rate of seed length,width and thickness slowed down.(2)There were significant differences in the contents of soluble sugar,crude starch and soluble protein in the seeds at different development stages(P<0.05).The contents of soluble sugar and crude starch showed unimodal curve changes,and the contents of soluble protein fluctuated in the range of 4.89%-5.82%.(3)There were significant differences in the content of reactive oxygen species and the activity of antioxidant enzymes(P<0.05).The H_(2)O_(2) content showed a unimodal curve with the-activities of POD and CAT,while the O_(2)^(-) content showed a similar trend with the activity of SOD.(4)There were significant differences in the contents of endogenous hormones in the seeds at different developmental stages(P<0.05).The contents of IAA,GA_(3) and ZR showed a trend of“first increase,then decrease and then increase”,while the ABA content showed a single-peak curve,and the ratio of(IAA+GA_(3)+ZR)/ABA showed a trend of“first decrease and then increase”.The minimum value was 1.001 at the 40th day after anthesis.【Conclusion】The activity of antioxidant enzymes and the content of endogenous hormones increased gradually from 10 to 30 days after anthesis,which effectively promoted the metabolism,division and differentiation of seed cells of bamboo.From 30 to 60 days after anthesis,the differentiation of bamboo seeds basically ended and the seeds entered the stage of nutrient accumulation,during which the activities of antioxidant enzymes and endogenous hormones decreased gradually,and the content of nutrients increased gradually.
