白藜芦醇抑制衰老小鼠脑组织小胶质细胞激活

Resveratrol inhibits the activation of microglia in the brain tissue of aging mice

摘要 目的探讨白藜芦醇(resveratrol,Res)对自然衰老小鼠认知能力的影响及其对小胶质细胞介导的炎症因子分泌的影响。方法选用2~3月龄和18~20月龄C57BL/6J小鼠为研究对象,分为三组,每组10只:青年对照组(Con);衰老模型组(Mod)和衰老给药组(Res)。Res组小鼠按45 mg/kg剂量每日灌胃白藜芦醇,连续8周。小鼠的学习记忆能力通过Morris水迷宫实验检测;小胶质细胞标记蛋白Ionized calcium binding adapter molecule(Iba-1)的表达变化通过免疫组织化学染色观察,并通过阳性细胞计数分析其表达情况。通过qRT-PCR技术检测炎症因子TNF-α和Arg-1基因的表达变化。结果通过Morris水迷宫实验结果显示,白藜芦醇治疗组小鼠学习记忆能力明显提高。免疫组织化学染色和体视学测量结果证明,白藜芦醇减少衰老小鼠脑组织中Iba-1的表达。qRT-PCR检测结果显示,白藜芦醇增加老年小鼠脑组织中Arg-1 mRNA的表达,减少TNF-αmRNA的表达。结论白藜芦醇改善老年小鼠的认知能力,其机制与抑制小胶质细胞介导的炎症反应有关。 Objective To explore the effect of resveratrol on the cognitive ability and the secretion of in-flammatory factors mediated by microglia in naturally aged mice.Methods C57BL/6J mice aged 2-3 months and 18-20 months were selected as the research subjects and divided into three groups,with 10 mice in each group:young control group(Con);aged control group(Mod)and aged dosing group(Res).Res group mice were given resveratrol by intragastric administration at a dose of 45mg/kg per day for eight consecutive weeks.Morris water maze experiment was performed to test the learning and memory ability of mice.The expression of Ionized calcium binding adapter molecule(Iba-1),the marker of microglia,was observed by immunohistochemical staining,and analyzed by positive cell count.The expression changes of TNF and Arg-1 were detected by qRT-PCR.Results The results of Morris water maze experiment showed that the learning and memory abilities of mice treated with resveratrol were significantly improved.Immunohistochemical staining and stereology measurement indicated that resveratrol reduced the expression of Iba-1 in brain.The results of qRT-PCR showed that resveratrol increased the expression of Arg-1 mRJVA and decreased the expression of TNF mRJVA in the brain of aged mice.Conclusions Resveratrol improves cognitive ability in elderly mice,and its mechanism is related to the inhibition of microglia mediated inflammatory responses.
