香芹酚和黄连素对小鼠子宫内膜炎炎性细胞因子及相关基因表达的影响

Effects of carvacrol and berberine on the expression of inflammatory cytokines and related genes in endometritis in mice

摘要 为了探究香芹酚和黄连素对小鼠子宫内膜炎炎性细胞因子及相关基因表达的影响,试验将64只小鼠随机分为4组,分别为空白组、模型组、黄连素治疗组和香芹酚治疗组,每组16只。空白组小鼠经子宫灌注生理盐水0.1 mL,模型组、黄连素治疗组和香芹酚治疗组子宫灌注大肠杆菌、金黄色葡萄球菌和无乳链球菌的混合菌液0.1 mL,连续5 d;第6天,空白组和模型组小鼠子宫灌注生理盐水,黄连素治疗组子宫灌注黄连素,香芹酚治疗组子宫灌注香芹酚,各0.1 mL,连续7 d;第13天处死所有小鼠,采集子宫组织,观察子宫组织病理学变化,采用ELISA试剂盒测定子宫组织中肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)和白细胞介素-8(IL-8)的质量浓度,采用实时荧光定量PCR法测定子宫中TNF-α、IL-1β、IL-6、IL-8、Toll样受体2(TLR2)和Toll样受体4(TLR4)的mRNA相对表达量。结果表明:模型组子宫内膜内陷,结缔组织排列疏松,黏膜上皮细胞脱落,炎性细胞增多,子宫组织中TNF-α、IL-1β、IL-6和IL-8的质量浓度均极显著高于空白组(P<0.01),TNF-α、IL-1β、IL-6、IL-8、TLR2和TLR4的mRNA相对表达量极显著高于空白组(P<0.01);黄连素组和香芹酚组子宫组织中TNF-α、IL-1β、IL-6和IL-8的质量浓度均极显著低于模型组(P<0.01),TNF-α、IL-1β、IL-6、IL-8、TLR2和TLR4的mRNA相对表达量极显著低于模型组(P<0.01)。除黄连素治疗组TNF-α质量浓度极显著高于香芹酚治疗组(P<0.01),TLR2 mRNA相对表达量显著高于香芹酚治疗组(P<0.05)外,其他指标两组间均差异不显著(P>0.05)。说明黄连素和香芹酚可以减轻大肠杆菌、金黄色葡萄球菌和无乳链球菌导致的小鼠子宫组织损伤,降低子宫组织中炎性细胞因子的质量浓度及相关基因的表达量,对子宫内膜炎具有治疗作用。 In order to explore the effects of carvacrol and berberine on the expression of inflammatory cytokines and related genes in endometritis in mice,in the experiment,sixty-four mice were randomly divided into four groups,namely blank group,model group,berberine treatment group and carvacrol treatment group,with 16 mice in each group.The blank group was intrauterine perfused with 0.1 mL of normal saline.The model group,berberine treatment group and carvacrol treatment group were intrauterine perfused with 0.1 mL of mixed bacterial solution of Escherichia coli,Staphylococcus aureus and Streptococcus agalactiae for 5 days,and on the 6th day,the control group and model group were perfused with normal saline,and berberine treatment group and carvacrol treatment group were intrauterine perfused with 0.1 mL of berberine and carvacrol respectively for 7 days.On the 13th day,all mice were sacrificed;uterine tissue was collected,and uterine histopathological changes were observed.The mass concentrations of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),interleukin-6(IL-6)and interleukin-8(IL-8)in uterine tissues were determined by ELISA kit.The relative mRNA expression levels of TNF-α,IL-1β,IL-6,IL-8,Toll-like receptor 2(TLR2)and Toll-like receptor 4(TLR4)in uterine tissue were determined by real-time quantitative PCR.The results showed that the endometrium was invaginated;the connective tissue was loose;the mucosal epithelial cells were shed,the inflammatory cells were increased;the mass concentrations of TNF-α,IL-1β,IL-6 and IL-8 in the uterine tissue were significantly higher than those in the blank group(P<0.01),and the relative mRNA expressions of TNF-α,IL-1β,IL-6,IL-8,TLR2 and TLR4 were also significantly higher than those in the blank group(P<0.01).The mass concentrations of TNF-α,IL-1β,IL-6 and IL-8 in uterine tissues of berberine group and carvacrol group were significantly lower than those in the model group(P<0.01),and the relative mRNA expressions of TNF-α,IL-1β,IL-6,IL-8,TLR2 and TLR4 were also significantly lower than those in the model group(P<0.01).Except that the concentration of TNF-αin the berberine treatment group were significantly higher than those in the carvacrol treatment group(P<0.01),and the relative expression of TLR2 mRNA were significantly higher than those in the carvacrol treatment group(P<0.05),there was no significant difference in other indexes between the two groups(P>0.05).These results indicated that berberine and carvacrol could alleviate the damage of mouse uterine tissues caused by Escherichia coli,Staphylococcus aureus and Streptococcus agalactiae,reduce the mass concentration of inflammatory cytokines in uterine tissue and the expression of related genes,and had therapeutic effects on endometritis.
