泡型棘球蚴病患者肝脏组织中LLT1表达观察

摘要 目的观察泡型棘球蚴病(AE)患者肝脏组织中凝集素样转录物1(LLT1)的表达变化,并分析其与AE临床病理参数的关系。方法收集30例AE患者的病灶肝脏组织样本,其中病灶近端肝脏组织(CLT,距离病灶中心<2 cm)30例、远端肝脏组织(DLT,距离病灶中心>2 cm)28例。采用免疫组化法观察LLT1的表达和定位。分析不同临床病理参数AE患者肝脏组织中LLT1的表达变化。从GEO数据库获取AE患者数据集GSE124362(包含6例CTL和6例匹配的DLT样本)分析LLT1编码基因CLEC2D与免疫细胞表达的相关性。结果LLT1主要表达于肝实质细胞和病灶周围的炎症细胞。CLT、DLT中LLT表达评分分别为3(2,4)、6(4,8)分,CLT中LLT1表达水平高于DLT(P<0.05)。LLT1表达与AE患者的P分期、ALT、AST和γ-谷氨酰转移酶水平相关(P均<0.05)。CLEC2D表达与浆细胞、M2型巨噬细胞和激活的树突状细胞呈正相关(r分别为0.83、0.87、0.74,P均<0.01),与激活的NK细胞、M1型巨噬细胞和肥大细胞呈负相关(r分别为-0.73、-0.60、-0.74,P均<0.01)。进一步分析发现,CLEC2D表达与M1型巨噬细胞标志CD1B和IL-6呈负相关(r分别为-0.65、-0.89,P均<0.05),与M2型巨噬细胞标志CD163、CLEC7A和IL-10呈正相关(r分别为0.90、0.71、0.74,P均<0.01)。结论LLT1在AE患者CLT中高表达,与患者的P分期和肝功能指标水平相关,与多种免疫细胞表达有关,LLT1可能参与AE病灶的浸润过程。 Objective To observe the alterations in lectin-like transcript 1(LLT1)expression within liver tissues of alveolar echinococcosis(AE)patients and to assess its correlation with clinical pathological parameters.Methods Thirty liver tissue samples from AE patients were collected,including 30 samples from proximal lesion tissues(CLT,<2 cm form the lesion center)and 28 samples from distal tissues(DLT,>2 cm from the lesion center).Immunohistochemistry was employed to determine the expression and localization of LLT1.The LLT1 expression changes in the liver tissues of AE patient with different clinical pathological parameters were analyzed.Data set GSE124362 from the GEO database,including 6 CTL and 6 matched DLT samples,was used to analyze the correlation between CLEC2D(the LLT1 coding gene)expression and immune cell expression.Results LLT1 predominantly expressed in hepatocytes and inflammatory cells within the lesions'surrounding inflammatory zone.The expression scores for LLT1 in CLT and DLT were 3(2,4)and 6(4,8),respectively,indicating significantly higher expression levels in CLT(both P<0.05).LLT1 expression was related to the AE patients'P stage,ALT,AST,andγ-glutamyl transferase levels(all P<0.05).CLEC2D expression showed a positive correlation with plasma cells,M2 macrophages,and activated dendritic cells(r=0.83,0.87,0.74,respectively;all P<0.01),and had a negative correlation with activated NK cells,M1 macrophages,and mast cells(r=–0.73,–0.60,–0.74,respectively;all P<0.01).Further analysis revealed that CLEC2D expression was negatively correlated with M1 macrophage markers CD1B and IL-6(r=–0.65,–0.89,respectively;both P<0.05),and was positively correlated with M2 macrophage markers CD163,CLEC7A,and IL-10(r=0.90,0.71,0.74,respectively;all P<0.01).Conclusion The high expression of LLT1 in CLT of AE patients was related to P stage,liver function indices,and various immune cell ex‐pression,suggesting LLT1 was involved in infiltration process of the AE liver lesions.
