摘要
研究天女木兰低温层积催芽种子中脱落酸含量变化规律,为解除休眠提供依据。以天女木兰种子为研究对象,采集低温沙藏层积0、30、60、90、120、150 d种子,采用高效液相色谱法测定种皮和胚乳(含胚)在低温层积过程中的脱落酸含量,采用石蜡切片法观测胚形态。结果表明:天女木兰0 d湿种子种皮和胚乳(含胚)中脱落酸含量明显高于0 d干种子;在低温沙藏层积过程中,种皮和胚乳(含胚)脱落酸含量随着处理时间的增加呈下降趋势;低温沙藏层积0~30 d胚乳(含胚)中的脱落酸含量高于种皮,但60 d后各处理种子胚乳(含胚)中的脱落酸含量均低于种皮;处理150 d,种皮与胚乳(含胚)中均无脱落酸。随着低温沙藏层积时间的增加,胚呈现逐渐增大的趋势,处理150 d胚的子叶变大。
Studying ABA change rule in the low temperature layered germination seeds of Magnolia sieboldii to provide basis for relieving seed dormancy.Taking the seeds of Magnolia sieboldii as the research object,the seeds were collected from 0,30,60,90,120 and 150 d low-temperature sand storage and layering processes.The changes of ABA content in the testa and endosperm(including embryo)of Magnolia sieboldii during low-temperature layering were measured by high-performance liquid chromatography.The results showed that the content of ABA in the seed coat and endosperm(including embryo)of 0 d wet seeds was significantly higher than that of 0 d dry seeds.The content of ABA in the seed coat and endosperm(including embryo)of Magnolia sieboldii seeds during low-temperature sand storage showed a decreasing trend with increasing of treatment time;The content of ABA in the endosperm(including embryo)of low-temperature sand storage layer 0~30 d was higher than that in the seed coat,but after 60 days,the content of ABA in the endosperm(including embryo)of all treated seeds was lower than that in the seed coat;At 150 days of treatment,there was no ABA in the seed coat and endosperm(including embryo);The embryo showed a gradually increasing trend with time increasing of low temperature sand storage layer,and the cotyledons of embryos treated for 150 days became larger.
