人乳头瘤病毒基因分型检测在绝经后妇女HPV感染中的应用价值

摘要 目的探究人乳头瘤病毒(HPV)基因分型检测在绝经后妇女HPV感染中的应用价值。方法选择80名接受HPV筛查的绝经后妇女为研究对象,采集其宫颈脱落细胞标本,采用聚合酶链式反应(PCR)-反向杂交法进行检测。以柱提法提取样本DNA,将提取液及阴、阳性对照品同时在天隆PCR扩增仪上进行扩增,在安必平杂交仪上进行杂交。分析阳性率、危险分型情况、阳性样本与感染样本分型情况及不同年龄段妇女HPV感染阳性率。结果在80名绝经后妇女中,HPV阳性检出率为15.00%(12/80)。其中,HPV-16型2例,占比16.67%;HPV-52型2例,占比16.67%;HPV-58型1例,占比8.33%;HPV-53型1例,占比8.33%;HPV-68型1例,占比8.33%;HPV-33型1例,占比8.33%;HPV-11型1例,占比8.33%;HPV-18型1例,占比8.33%;HPV-cp8304型1例,占比8.33%;HPV-66型1例,占比8.33%。在12例HPV阳性样本中,多重感染样本3例,占比25.00%;单一感染样本9例,占比75.00%。不同年龄组HPV感染阳性例数分布数据差异不明显(P>0.05)。结论绝经后不同年龄段妇女HPV阳性率无明显分布,感染以单一感染为主,HPV基因分型检测有着较高的检验诊断价值,值得推广。 Objective To explore the application value of human papillomavirus(HPV)genotyping in postmenopausal women with HPV infection.Methods 80 postmenopausal women who underwent HPV screening were selected as the study subjects,and their cervical exfoliated cell samples were collected and detected by polymerase chain reaction(PCR)-reverse hybridization.The DNA was extracted by column extraction,and the extracted solution,negative and positive control products were amplified by Tianlong PCR and hybridized by Ambipine hybridization.The positive rate,risk typing,typing of positive and infected samples,and positive rate of HPV infection in women of different ages were analyzed.Results Among 80 postmenopausal women,the HPV positive rate was 15.00%(12/80).Among them,HPV-16 type 2 cases,accounting for 16.67%;HPV-52 type 2 cases,accounting for 16.67%;HPV-58 type 1 case,accounting for 8.33%;HPV-53 type 1 case,accounting for 8.33%;HPV-68 type 1 case,accounting for 8.33%;HPV-33 type 1 case,accounting for 8.33%;HPV-11 type 1 case,accounting for 8.33%;HPV-18 type 1 case,accounting for 8.33%;HPV-cp8304 was found in 1 case,accounting for 8.33%.HPV-66 type 1 case,accounting for 8.33%.Among the 12 HPV-positive samples,3 were multiple infected,accounting for 25.00%;9 were single infected,accounting for 75.00%.There was no significant difference in the distribution of HPV positive cases in different age groups(P>0.05).Conclusion There was no obvious distribution of HPV positive rate in postmenopausal women of different ages,and the infection was mainly single infection.HPV genotyping has a high diagnostic value and is worth promoting.
