摘要
目的:目前免疫组织化学染色阳性对照的制作方法多种多样,单组织对照制作简单,但使用时由于免疫组织化学染色指标过多,容易出错。一些多组织对照使用方便,但制作方法繁琐、难度高。本研究旨在探讨一种新型、简单的多组织蜡块的制作及其在免疫组织化学染色对照中的应用。方法:依据国际特设专家委员会对常见抗体对照组织的推荐,选取包含肝、阑尾、扁桃体、肺、胰腺组织蜡块各1个为1组,共10组,进行厚度为6μm的连续切片,将切好的组织片依次卷曲叠在一起,形成一个长条形的致密组织卷,经过切割和包埋,制成多组织免疫组织化学染色阳性对照蜡块,然后用于免疫组织化学染色。结果:该阳性对照蜡块染色效果好,包含5种阳性对照组织,可适用于68种常用的免疫组织化学染色标志物。一次至少可制作对照蜡块2个,耗时约20 min,组织大小平均约为4.7 mm×4.1 mm,不同对照蜡块之间以及同一对照蜡块的不同切面之间,切片后免疫组织化学染色结果一致性较好,一个蜡块平均可切约800张有效的免疫组织化学染色对照片。结论:多组织阳性对照蜡块制作方法简单,耗时少、组织面积小、一致性好、重复性高,可以满足68种常用抗体对照组织的设置需求,对日常免疫组织化学染色检测有较好的临床应用价值。
Objective:Currently,there are various methods for producing positive controls for immunohistochemical staining.Single-tissue controls are simple to make,but their use can lead to errors due to the multitude of immunohistochemical staining indicators.Some multitissue controls are convenient to use but are complex and difficult to produce.This study aims to explore a novel and simple method for creating multi-tissue paraffin blocks and their application in immunohistochemical staining controls.Methods:Based on the recommendations of the International Ad Hoc Expert Committee for common antibody control tissues,paraffin blocks containing liver,appendix,tonsil,lung,and pancreas tissues were selected,with one block from each tissue type forming a group.Ten groups were prepared,and continuous sections of 6μm thickness were cut.The tissue sections were then rolled and stacked together to form a long,dense tissue roll,which was subsequently cut and embedded to create multi-tissue immunohistochemical staining positive control paraffin blocks.These blocks were then used in immunohistochemical staining.Results:The positive control paraffin blocks demonstrated good staining effects,containing 5 types of positive control tissues and suitable for 68 commonly used immunohistochemical staining markers.At least 2 control paraffin blocks were made at a time,taking approximately 20 minutes.The average tissue size was about 4.7 mm×4.1 mm.The immunohistochemical staining results were consistent between different control blocks and different sections of the same control block.On average,one paraffin block yielded approximately 800 effective immunohistochemical staining sections.Conclusion:The multi-tissue positive control paraffin blocks are simple to produce,timeefficient,have small tissue areas,good consistency,and high repeatability.They can meet the needs for setting up controls for 68 commonly used antibodies and have good clinical application value for routine immunohistochemical staining.
