半夏与清半夏中水溶性成分的含量测定

摘要 文章建立了能够同时测定半夏中尿嘧啶、次黄嘌呤、黄嘌呤、尿苷、肌苷、腺苷6种化学成分,以及清半夏中次黄嘌呤、尿苷、肌苷、鸟苷、腺苷5种成分含量的方法。文章采用高效液相色谱法,选择色谱柱为依利特ODS2(4.6 mm×250 mm,5μm),流动相为甲醇-0.1%甲酸水,梯度洗脱,检测波长为265 nm,对半夏及清半夏中的水溶性成分进行检测,并通过回归方程计算半夏中6种水溶性成分的含量,以及清半夏中5种水溶性成分的含量。结果显示,半夏中的6种成分及清半夏中的5种成分呈良好线性关系。除鸟苷外,清半夏与生半夏相比,其中核苷类成分的含量明显降低,而鸟苷的含量有所增加;炮制后的清半夏中测得的水溶性成分的含量均比生半夏要低,且半夏中的尿嘧啶与黄嘌呤两种成分消失或转化成了其他成分。文章所建立的方法可以一次性检测半夏中的6种水溶性成分和清半夏中的5种水溶性成分,具有简便、灵敏、可控的特点,可作为测定半夏饮片及清半夏中水溶性成分含量的有效方法。 The metod which can analyze 6 kinds of chemical constituents,such as urinary pyrimidine,Xanthine,xanthine,urinary glycosides,inosine and adenosine in Banxia,and the 5 components of Middle xanthine,urinary glycosides,inosine,guanosine and adenosine in Qingbanxia has been established.High Performance liquid chromatography(HPLC)was used to analyze the water-soluble components in Banxia and in Qingbanxia,whose chromatographic column was Elit ODS2(4.6 mm×250 mm,5μm),flow phase was methanol-0.1%formic acid water,gradient elution,detection wavelength 265 nm.The content of 6 water-soluble components in Banxia and the content of 5 kinds of water-soluble components in the Qingbanxia were calculated by regression equation.The 6 components in Banxia and the 5 components in Qingbanxia showed a good linear relationship.In addition to guanosine,the content of nucleoside components in Qingbanxia was significantly reduced compared with Banxia;the content of guanosine in Qingbanxia increased,and the content of water-soluble components determined in Qingbanxia after concoction was lower than that in Banxia.The content of water-soluble components measured in the finished Qingbanxia was lower than that in the middle half of the Banxia,and the contents of the urine pyrimidine and xanthine in the Banxia were disappeared or converted into other components.This method can detect 6 water-soluble components in Banxia and 5 water-soluble components in Qingbanxia,which is simple,sensitive and controllable,and can determine the content of water-soluble components in Banxia decoction tablets and Qingbanxia.
