低共熔溶剂提取晋产吸水石栽培铁皮石斛多糖工艺优化及抗氧化活性研究

Optimization of extraction process of polysaccharides from Dendrobium officinale cultivated on water-absorbing stone in Shanxi Province using low eutectic solvents and its antioxidant activity

摘要 铁皮石斛为我国传统的药用和食用同源的植物,具有极高的中药价值,尤其是其多糖含量最为显著。本研究以山西产吸水石栽培的铁皮石斛为原材料,通过单因素及响应面分析法优化低共熔溶剂提取铁皮石斛多糖工艺,然后分析不同浓度多糖对DPPH自由基、羟基自由基和ABTS+自由基的清除效果。结果显示,低共熔溶剂提取铁皮石斛多糖工艺:采用L-脯氨酸-草酸低共熔溶剂体系,在料液比1∶21(g/mL)、温度56℃的条件下超声30 min,最终多糖得率为(34.35±0.2)%。进行抗氧化实验分析,发现5 mg/mL的多糖浓度对DPPH自由基、羟基自由基和ABTS自由基的清除率最高,达到37.7%、96.6%和77.8%。本研究为铁皮石斛多糖的提取提供了一种有效的方法,同时探究了多糖添加量对抗氧化能力的变化,为铁皮石斛多糖的提取和抗氧化添加剂的开发提供思路。 Dendrobium officinale is a traditional plant in China used for both medicinal and edible purposes,with high traditional Chinese medicinal value,especially due to its significant polysaccharide content.Dendrobium officinale cultivated on water-absorbing stones in Shanxi province was taken as the raw materials,the polysaccharide extraction process was optimized using eutectic solvent extraction technique through single-factor and response surface analysis.Subsequently,the scavenging effects of different concentrations of polysaccharides on DPPH,hydroxyl radicals,and ABTS+were analyzed.The results showed that the optimized polysaccharide extraction process of Dendrobium officinale using a L-proline-oxalic acid eutectic solvent system was as follows:with a material-to-liquid ratio of 1∶21(g/mL),the temperature of 56℃,under ultrasound for 30 minutes,resulted in a polysaccharide yield of(34.35±0.2)%.Antioxidant experiments revealed that a polysaccharide concentration of 5 mg/mL exhibited the highest scavenging rates for DPPH,hydroxyl radicals,and ABTS+,reaching 37.7%,96.6%,and 77.8%,respectively.This study provides an effective method for the extraction of polysaccharides from Dendrobium officinale,and explores the impact of polysaccharide concentration on antioxidant capacity,offering insights for the extraction of polysaccharides from Dendrobium officinale and the development of antioxidant additives.
