摘要
铜(Cu)是参与体内多种生物代谢途径的辅助因子,在细胞呼吸、神经递质传递等生理过程中发挥重要作用。铜死亡的关键在于Cu离子蓄积并结合三羧酸循环脂酰化组分,导致脂酰化蛋白聚集和铁-硫簇蛋白损失,形成细胞蛋白毒性应激。金属Cu参与下的铜死亡与肿瘤疾病的形成、发展具有密切关系。目前研究肿瘤病因、进展机制主要是基于自噬、凋亡、焦亡、铁死亡等方面,研究肿瘤微环境中上皮-间质转化、细胞外基质重塑及血管内皮生长因子表达等。目前,中医药对铜死亡的实验研究尚不足,因此,结合中医药研究铜死亡及其抗肿瘤机制尚有很大探索空间。
Copper(Cu)is a cofactor involved in a variety of biological metabolic pathways in the body and plays an important role in physiological processes such as cellular respiration and neurotransmitter transmission.The key to cuproptosis is the accumulation of Cu ions combined with the fatty acylated components of the tricarboxylic acid cycle,leading to the aggregation of fatty acylated proteins and the loss of iron-sulfur cluster proteins,thus resulting in cellular proteotoxic stress.cuproptosis is closely related to the formation and development of tumor diseases.At present,the etiology and progression mechanism of tumors are mainly based on autophagy,apoptosis,pyroptosis,Ferroptosis and other aspects.In addition,the epithelial-mesenchymal transition,extracellular matrix remodeling and vascular endothelial growth factor expression in tumor microenvironment are studied.The experimental study of effect on cuproptosis of TCM intervention is insufficient,so there is still a lot of space for exploring cuproptosis and its anti-tumor mechanism with traditional Chinese medicine.
