摘要
目的采用RP-HPLC-UV同时测定定喘止嗽丸中3种罂粟壳生物碱(吗啡、可待因、罂粟碱)的含量。方法采用Kromasil100-2.5 C_(18)色谱柱(250 mm×4.6 mm,5μm),以乙腈-0.1%磷酸水为流动相,梯度洗脱;流速为1.0 mL·min^(-1);柱温:30℃;检测波长为210 nm(吗啡、可待因)和251 nm(罂粟碱)。结果吗啡、磷酸可待因和盐酸罂粟碱分别在0.2957~9.8561μg·mL^(-1)(R^(2)=0.9999)、0.0729~2.4298μg·mL^(-1)(R^(2)=0.9998)、0.0464~1.4730μg·mL^(-1)(R^(2)=0.9998)内线性关系良好;平均加样回收率为89.5%~91.5%,RSD为1.10%~3.91%。结论该方法精密稳定,重复性好,可用于定喘止嗽丸的质量控制。
OBJECTIVE To establish an RP-HPLC-UV method for the simultaneous contentdeter mination of morphine,codeine,papaverine from Opium Poppy Shell in Dingchuanzhisou pills.METHODS The separation was performed on a Kromasil 100-2.5 C_(18) column(250 mm×4.6 mm,5μm)with the mobile phase comprising of acetonitrile-0.1%phosphoric acid flowing at 1.0 mL·min^(-1)in a gradient elution manner,the column temperature was 30℃and the detection wave length was set at 210 nm(morphine,codeine)and 251 nm(papaverine).RESULTS Good linear relationship was observed for morphine,codeine phosphate and papaverine hydrochloride in the range of 0.2957-9.8561μg·mL^(-1)(R^(2)=0.9999),0.0729-2.4298μg·mL^(-1)(R^(2)=0.9998),and 0.0464-1.4730μg·mL^(-1)(R^(2)=0.9998),respectively.The average recoveries were 89.5%-91.5%with the RSD values of 1.10%-3.91%.CONCLUSION This precise stable and reproducible method can be used for the quality control of Dingchuanzhisou pills.
