摘要
目的探究三阴性乳腺癌(TNBC)组织中微RNA-331-3p(miR-331-3p)和跨膜蛋白119(TMEM119)水平变化与临床预后的相关性。方法选取2017年1月至2019年1月在信阳市中心医院行手术治疗的98例TNBC病人的癌组织以及癌旁组织,分别作为TNBC组和癌旁组。在术后3年对所有病人进行随访,分为预后良好组为23例,预后不良组为75例。通过实时荧光定量PCR(qRT-PCR)法检测miR-331-3p和TMEM119的表达水平;采用Pearson法分析TNBC组织中miR-331-3p和TMEM119表达水平的相关性;分析miR-331-3p和TMEM119水平与TNBC病人预后的关系;影响TNBC病人预后的影响因素采用Cox回归的方法进行分析。结果TNBC组TMEM119(1.53±0.24)的表达水平与癌旁组相比(1.01±0.13)显著上升,miR-331-3p(0.74±0.13)的表达水平与癌旁组(1.02±0.11)相比显著下降(t=41.53、41.53,P<0.05)。Pearson相关性分析结果显示,miR-331-3p和TMEM119表达水平呈负相关关系(r=−0.49,P<0.001)。miR-331-3p和TMEM119表达水平与肿瘤长径、TNM分期、腋窝淋巴结转移有关(P<0.05)。miR-331-3p高表达组病人3年内生存率16/49(32.65%)高于低表达组7/49(14.29%);TMEM119高表达组病人3年内生存率4/49(8.16%)低于低表达组19/49(38.78%)(Z=4.60,P=0.032;Z=12.78,P<0.001)。预后不良组miR-331-3p(0.65±0.13)表达水平与预后良好组(1.03±0.12)相比显著降低,TMEM119(1.69±0.27)表达水平与预后良好组(1.02±0.14)相比显著升高(t=38.74、33.30,P<0.05)。结果显示,TNM分期(HR=5.88)、miR-331-3p(HR=2.45)和TMEM119(HR=2.86)表达水平是影响TNBC病人预后的危险因素(P<0.05)。结论TNBC病人癌组织中miR-331-3p低表达,TMEM119高表达,二者水平与预后评估密切相关。
Objective To explore the correlation between the changes of microRNA-331-3p s(miR-331-3p)and transmembrane protein 119(TMEM119)levels in triple negative breast cancer(TNBC)tissues and clinical prognosis.Methods The cancerous tissues and adjacent tissues of 98 patients with TNBC who underwent surgical treatment in Xinyang Central Hospital from January 2017 to January 2019 were selected as TNBC group and adjacent group respectively.All patients were followed up 3 years after surgery,and were divided into good prognosis group(23 cases)and bad prognosis group(75 cases).The expression levels of miR-331-3p and TMEM119 were detected by real-time fluorescent quantitative PCR(qRT-PCR);Pearson method was used to analyze the correlation between miR-331-3p and TMEM119 expression levels in TNBC tissues;The relationship between the levels of miR-331-3p and TMEM119 and the prognosis of TNBC patients was analyzed.The factors influencing the prognosis of TNBC patients were analyzed by Cox regression method.Results The expression level of TMEM119(1.53±0.24)in the TNBC group was significantly increased compared with that in the para-cancer group(1.01±0.13),and the expression level of miR-331-3p(0.74±0.13)was significantly decreased compared with that in the para-cancer group(1.02±0.11)(t=41.53,41.53,P<0.05).Pearson correlation analysis showed that there was a negative correlation between miR-331-3p and TMEM119 expression levels(r=−0.49,P<0.001).The 3-year survival rate of patients with high expression of miR-331-3p was 16/49(32.65%)higher than that of patients with low expression of Mir-331-3p was 7/49(14.29%).The 3-year survival rate of TMEM119 patients in the high expression group was 4/49(8.16%)lower than that in the low expression group 19/49(38.78%)(Z=4.60,P=0.032;Z=12.78,P<0.001).The expression level of miR-331-3p(0.65±0.13)in the poor prognosis group was significantly lower than that in the good prognosis group(1.03±0.12),and the expression level of TMEM119(1.69±0.27)was significantly higher than that in the good prognosis group(1.02±0.14)(t=38.74,33.30,P<0.05).The results showed that the expression levels of TNM stage(HR=5.88),miR-331-3p(HR=2.45)and TMEM119(HR=2.86)were risk factors affecting the prognosis of TNBC patients(P<0.05).Con clusion The expression of miR-331-3p is low and TMEM119 is high in cancer tissues of TNBC patients,and the levels of both are closely related to prognosis assessment.
