摘要
对实验室保藏的几株芽孢杆菌进行耐受性试验,并探究其产酶条件。结果表明:芽孢杆菌最适生长pH范围为6.0~8.0,最适生长温度为35℃,最佳培养转速为150~180 r/min。菌株在固态培养基中生长最适水分条件为35%,水分大于35%时蛋白酶活性较高,蛋白酶活力最高为143.63 U/g。温度对芽孢杆菌的生长影响较为明显,在35~45℃范围内生长较好,最佳产蛋白酶温度为40℃。酸度对芽孢杆菌生长影响大,在pH5~6条件下对菌株产生不同程度抑制,菌株在酸性和偏中性条件下均能产生蛋白酶。正交试验表明,芽孢杆菌在40%含水率、35℃、pH8.0固态培养条件下产蛋白酶活力最高。
This study conducted tolerance tests on several strains of Bacillus stored in the laboratory and explored their enzyme production conditions.The results showed that the optimal pH range for Bacillus growth was 6.0-8.0,the optimal growth temperature was 35℃,and the optimal cultivation speed was 150-180 rpm.The optimal water condition for the growth of the strain in solid-state culture medium is 35%.When the water content is greater than 35%,the protease activity is higher,and the highest protease activity is 143.63 U/g.The temperature has a significant impact on the growth of Bacillus,with better growth in the range of 35-45℃.The optimal temperature for protease production is 40℃.Acidity has a significant impact on the growth of Bacillus.Under pH 5-6 conditions,the strain is inhibited to varying degrees.The strain can produce protease under both acidic and neutral conditions.The orthogonal experiment showed that Bacillus had the highest protease activity under solid-state cultivation conditions of 40%water content,35℃,and pH 8.0.
