摘要
目的研究九味楮实颗粒在治疗雌性中枢性性早熟过程中对大鼠甲状腺激素水平及甲状腺组织形态的影响。方法48只23日龄雌性SD大鼠,按照随机表法分成对照组、模型组、西药组和中药低、中、高剂量组,每组8只。采用N-甲基-DL-天冬氨酸(NMDA)建立大鼠性早熟模型,中药低、中、高剂量组分别选用3种倍增剂量(17.37 g/kg、34.74 g/kg和69.48 g/kg)的九味楮实颗粒干预;西药组用戈那瑞林干预。观察大鼠的饮水、进食和活动情况以及甲状腺滤泡细胞形态;采用ELISA试剂盒法检测大鼠血清中卵泡刺激素(FSH)、促黄体生成素(LH)、雌二醇(E_(2))、游离甲状腺素(FT_(3、)FT_(4))及促甲状腺激素(TSH)水平。结果与对照组比较,模型组大鼠血清LH、FSH、E_(2)水平升高,TSH水平降低,FT_(3)、FT_(4)水平升高(P<0.05)。与模型组比较,西药组、中药中、高剂量组大鼠血清LH、FSH、E_(2)水平降低,TSH水平升高,FT_(4)水平降低,FT_(3)水平轻度降低(P<0.05);中药低剂量组较模型组FSH水平降低,FT4水平降低(P<0.05)。结论九味楮实颗粒在治疗性早熟过程中可能影响FT_(3)、FT_(4)、TSH的水平。
Objective To investigate the effect of Jiuwei Chushi Granules on the thyroid hormone level and thyroid tissue morphology in female central precocious puberty rats during the treatment of precocious puberty.Methods 48 female SD rats at the age of 23 days were randomly divided into control group,model group,Western medicine group,TCM low,medium and high dose groups,with 8 rats in each group.The rat model of precocious puberty was established with N-methyl-DL-aspartic acid(NMDA).TCM low,medium and high dose groups were treated with three kinds of double doses(17.37 g/kg,34.74 g/kg and 69.48 g/kg)of Jiuwei Chushi Granules;Western medicine group was intervened by gonnarelin.The drinking,eating and activity of rats and the morphology of thyroid follicular cells were observed;the serum levels of follicle-stimulating hormone(FSH),luteinizing hormone(LH),estradiol(E_(2)),free thyroxine(FT_(3),FT_(4))and thyroid stimulating hormone(TSH)were measured by ELISA.Results Compared with control group,serum LH,FSH and E_(2) levels in model group increased,TSH levels decreased,FT_(3) and FT_(4)levels increased(P<0.05);compared with model group,serum LH,FSH and E_(2) levels in Western medicine group,TCM middle and high dose groups decreased,TSH levels increased,FT_(4) levels decreased,and FT_(3) levels slightly decreased(P<0.05);compared with model group,FSH and FT_(4) levels in TCM low dose group were lower(P<0.05).Conclusion Jiuwei Chushi Granules may affect FT_(3),FT_(4) and TSH levels in treatment of precocious puberty.
