一株鸭源多杀性巴氏杆菌的分离鉴定及耐药性分析

摘要 为确认山东省德州市某肉鸭场疑似为鸭多杀性巴氏杆菌感染的病原菌,本试验对于送检病死鸭进行剖检、细菌分离培养、菌落观察、生化试验、PCR鉴定、16Sr RNA基因测序分析以及药敏试验等方法鉴定该病原菌。结果显示:分离得到的病原菌在含有5%胎牛血清的营养琼脂培养基中呈现为圆形、湿润、光滑的灰白色菌落,革兰氏染色为阴性短杆菌,瑞氏染色呈现两极浓染;分离菌株的生化特性可以发酵葡萄糖、蔗糖以及甘露醇,硫化氢、氧化酶等试验阳性;细菌特异性PCR扩增片段与预期条带大小一致;16S rRNA基因序列系统进化树显示与多杀性巴氏杆菌同源性为99%;分离菌株对于新霉素、阿莫西林、氨苄西林、头孢噻肟、林可霉素具有耐药性,对于氟苯尼考、恩诺沙星、多西环素、粘杆菌素B、卡那霉素、大观霉素敏感。本研究结果为鸭源多杀性巴氏杆菌病的防治提供一定参考依据。 In order to investigate the suspected pathogenic bacteria causing Pasteurella multocida infection in ducks at a meat-type duck farm in Dezhou,Shandong Province,various methods were employed.These methods included necropsy,bacterial isolation and culture,colony observation,biochemical experiments,PCR identification,16S rRNA gene sequencing analysis,and drug sensitivity testing of deceased ducks.The findings revealed that the isolated pathogenic bacteria cultivated on nutrient agar medium supplemented with 5%fetal bovine serum exhibited characteristics of round,slightly raised,moist,smooth,and translucent colonies.The Gram staining results indicated a negative presence of Brevibacterium,while the Wright staining revealed poles staining.The isolated strains exhibited the ability to ferment glucose,sucrose,and mannitol,and yielded positive results for hydrogen sulfide,oxidase,and other tests.The amplified fragment obtained through bacterial specific PCR was found to be consistent with the expected band size.The phylogenetic analysis of the 16S rRNA gene sequence demonstrated a 99%homology with Pasteurella multocida.Furthermore,the isolated strains displayed resistance to neomycin,amoxicillin,ampicillin,cefotaxime,and lincomycin,while exhibiting sensitivity to florfenicol,enrofloxacin,and doxycycline,colistin B,kanamycin,spectinomycin.The findings of this study offer valuable insights for the prevention and treatment of Pasteurella multocida in ducks.
