摘要
目的 观察脾胃培源方对慢性萎缩性胃炎(CAG)模型大鼠胃黏膜炎症及环氧合酶-2(COX-2)mRNA和蛋白表达的影响,探讨脾胃培源方治疗CAG的可能作用机制。方法 从54只SPF级健康雄性SD大鼠中随机抽取8只作为正常组,其余大鼠通过饮用氨水联合脱氧胆酸钠溶液灌胃的方法制备CAG大鼠模型,10周后,将模型复制成功的40只大鼠采用随机数字表法分为模型组,胃复春组,脾胃培源方高、中、低剂量组,每组8只。各组予相应药物或生理盐水连续灌胃4周后,比较各组大鼠体质量增长情况;取大鼠胃组织,镜下观察大鼠胃黏膜病理学改变,并分别采用蛋白免疫印迹法、免疫组化法及聚合酶链式反应3种方法检测大鼠胃组织中COX-2蛋白及mRNA的表达情况。结果 与正常组比较,模型组大鼠体质量增长率明显降低,差异有统计学意义(P<0.01);与模型组比较,脾胃培源方各剂量组大鼠体质量增长率明显升高,差异均有统计学意义(P<0.01)。与正常组比较,模型组大鼠胃组织中COX-2蛋白及mRNA表达水平明显升高,差异均有统计学意义(P<0.01);与模型组比较,脾胃培源方各剂量组大鼠胃组织中COX-2蛋白及mRNA表达水平明显降低,差异均有统计学意义(P<0.01)。模型组大鼠胃黏膜变薄,固有腺体萎缩,炎性细胞全层浸润;脾胃培源方可明显改善及修复大鼠胃黏膜,减轻胃黏膜损伤。结论 脾胃培源方可促进CAG模型大鼠体质量增长,改善及修复大鼠胃黏膜,作用机制与其抑制COX-2的过度表达密切相关。
Objective To investigate the impact of Piwei Peiyuan Fang(脾胃培源方)on gastric mucosal inflammation and cyclooxygenase-2(COX-2)mRNA and protein expression in rats with chronic atrophic gastritis(CAG),and to explore the potential mechanism of Piwei Peiyuan Fang in treating CAG.Methods 8 out of 54 SPF healthy male SD rats were randomly assigned to the normal group,while the remaining rats were induced with CAG models by drinking ammonia and intragastric administration of deoxycholic acid sodium solution.After 10 weeks,40 successfully modeled rats were divided into model group,Weifuchun group,as well as high-dose,medium-dose and low-dose groups of Piwei Peiyuan Fang using a random number table method,each consisting of 8 rats.Following 4 weeks of continuous intragastric administration of corresponding drugs or normal saline,body mass growth was compared among the groups.Pathological changes in gastric mucosa were observed under a microscope.The expression levels of COX-2 protein and mRNA in gastric tissue were detected using western blotting,immunohistochemistry and polymerase chain reaction.Results Compared with normal group,the growth rate of body mass of rats in model group decreased significantly,the difference was statistically significant(P<0.01).Compared with model group,the growth rate of body mass of rats in each dose group of Piwei Peiyuan Fang was significantly increased,and the differences were statistically significant(P<0.01).Compared with normal group,the expression levels of COX-2 protein and mRNA in gastric tissues of rats in model group were significantly increased,with statistical significance(P<0.01).Compared with model group,the expression levels of COX-2 protein and mRNA in gastric tissues of rats in each dose group of Piwei Peiyuan Fang were significantly decreased,and the differences were statistically significant(P<0.01).In the model group,gastric mucosa became thin,intrinsic glands atrophied,and inflammatory cells infiltrated the whole layer.Piwei Peiyuan Fang can obviously improve and repair the gastric mucosa of rats and reduce the damage of gastric mucosa.Conclusion Piwei Peiyuan Fang can promote the body mass growth of CAG model rats,improve and repair the gastric mucosa of rats,and the mechanism is closely related to the inhibition of COX-2 over-expression.
