NAC6转录因子在芒果果实采后类胡萝卜素代谢过程中的表达及影响

摘要 NAC转录因子在植物生长发育过程中具有重要调节作用。为了探明NAC6转录因子对芒果果实采后类胡萝卜素代谢的影响作用,选取‘台农’和‘金煌’2个品种芒果商业成熟时期果实作为实验材料,在恒温(20℃)、恒湿(相对湿度85%)、避光环境进行贮藏处理,并测定果皮、果肉中类胡萝卜素组分含量变化、NAC6转录因子及类胡萝卜素代谢关键酶基因表达。结果表明:2个品种芒果果实均含有α-胡萝卜素、叶黄素、β-胡萝卜素、β-隐黄素和玉米黄素;在整个贮藏过程中,NAC6转录因子在2个品种芒果果实中的表达均呈持续上升的趋势,在贮藏后期达到极大值,随后略有下降;相关性分析表明,NAC6转录因子可能诱导了芒果果实中1-脱氧-木酮糖-5-磷酸异构酶、法尼基焦磷酸合成酶、牻牛儿基牻牛儿基焦磷酸合成酶、八氢番茄红素合成酶、八氢番茄红素脱氢酶、ζ-胡萝卜素脱氢酶、类胡萝卜素异构酶、β-胡萝卜素羟化酶及紫黄素脱环氧化酶基因的表达,抑制了番茄红素β-环化酶、新黄素合成酶及9-顺式环氧类胡萝卜素双加氧酶基因的表达;NAC6转录因子在‘台农’芒果果皮、果肉中的表达与类胡萝卜素裂解双加氧酶基因(carotenoid cleavage dioxygenase,CCD)的表达呈显著负相关,相关系数分别为-0.8410、-0.8383,NAC6转录因子可能通过抑制CCD的表达,从而延缓果实类胡萝卜素的降解速率。本研究结果能为芒果果实采后贮藏保鲜及品质调控技术提供依据。 The NAC transcription factors play an important regulatory role in plant growth and development.In order to explore the impact of NAC6 on carotenoid metabolism in postharvest mango fruit,this study investigated changes in the carotenoid contents as well as the expression of NAC6 and key enzymes involved in carotenoid synthesis and metabolism in the mature peel and pulp of 2 mango varieties,“Tainong”and“Jinhuang”,at commercial maturity,during postharvest storage under constant temperature(20℃),constant humidity(85%),and dark conditions.The results showed that both mango varieties containedα-carotene,lutein,β-carotene,β-cryptoxanthin,and zeaxanthin.Throughout the storage process,the expression of NAC6 in both mango varieties showed a continuous upward trend,reaching a peak in the late stage of storage,followed by a slight decline.Correlation analysis showed that NAC6 might induce the gene expression of 1-deoxy-D-xylulose-5-phosphate reductoisomerase,farnesyl pyrophosphate synthase,geranylgeranyl pyrophosphate synthase,phytoene synthase,phytoene desaturase,ζ-carotene desaturase,carotenoid isomerase,β-carotene hydroxylase,and violaxanthin de-epoxidase in mango fruit,while inhibiting the gene expression of lycopeneβ-cyclase,neoxanthin synthase,and 9-cis-epoxycarotenoid dioxygenase.The expression of NAC6 in the peel and pulp of“Tainong”mango was significantly negatively correlated with the gene expression of carotenoid cleavage dioxygenase(CCD),with correlation coefficients of-0.8410 and-0.8383,respectively.NAC6 might delay the degradation of carotenoids by inhibiting the expression of CCD.The results from this research provide an experimental basis for the postharvest storage and preservation of mango fruit as well as its quality control.
