混合大孔树脂纯化蓝莓花青素及抗氧化性研究

摘要 研究混合大孔树脂对蓝莓花青素的纯化工艺条件,提高蓝莓花青素的抗氧化效果。比较5种大孔树脂D101型、HP-20型、AB-8型、NKA-9型、D3520型对蓝莓粗提液的吸附效果。以吸附率和解吸率为评价指标,通过单因素试验和正交试验优化混合树脂分离纯化蓝莓花青素的吸附和解吸条件,分析纯化前后蓝莓花青素的抗氧化性,对比纯化前后花青素抗羟基自由基(·OH)和2,2-二苯基-1-苦基肼自由基(DPPH·)的差异。结果表明,D101型、HP-20型两种树脂按9∶1混合,混合树脂与蓝莓粗提液按1∶1吸附效果较好;当超声频率为40 kHz、吸附样液浓度为原样液、吸附时间为40 min、吸附温度为20℃时,吸附率达到96.2%;当上样量为5 mL、上样浓度为原样液、洗脱流速为1.0 mL/min、洗脱剂乙醇浓度为60%、洗脱剂体积为3 BV时,解吸率高达95.7%。采用混合树脂纯化的蓝莓花青素抗·OH和DPPH·均高于纯化前。纯化前后花青素对·OH的清除率分别为96.25%和97.85%,纯化前后对DPPH·的清除率分别为88.35%和93.20%。D101型、HP-20型混合树脂对蓝莓花青素的纯化效果良好,可用于蓝莓花青素的纯化;纯化后的蓝莓花青素抗氧化能力明显提高,能更好地清除·OH和DPPH·,且对·OH的清除效果高于DPPH·。 Purification process conditions of blueberry anthocyanins by mixed macroporous resins are studied to improve the antioxidant effect of blueberry anthocyanins.The adsorption effects of five macroporous resins such as D101,HP-20,AB-8,NKA-9 and D3520 on blueberry crude extract are compared.With adsorption rate and desorption rate as the evaluation indexes,the adsorption and desorption conditions for the separation and purification of blueberry anthocyanins by mixed resins are optimized through single factor test and orthogonal test.The antioxidant properties of blueberry anthocyanins before and after purification are analyzed,and the differences in hydroxyl free radicals(·OH)and 2,2-diphenyl-1-picrylhydrazine free radicals(DPPH·)scavenging rates of anthocyanins before and after purification are compared.The results show that D101 and HP-20 resins are mixed in a ratio of 9∶1,and the adsorption effect is better when the ratio of the mixed resins to blueberry crude extract is 1∶1.When the ultrasonic frequency is 40 kHz,the concentration of adsorption sample solution is the original sample solution,the adsorption time is 40 min,and the adsorption temperature is 20℃,the adsorption rate reaches 96.2%.When the loading volume is 5 mL,the loading concentration is the original sample solution,the elution velocity is 1.0 mL/min,the eluent ethanol concentration is 60%,and the eluent volume is 3 BV,the desorption rate is as high as 95.7%.The·OH and DPPH·scavenging rates of blueberry anthocyanins purified with mixed resins are higher than those before purification.The scavenging rates of anthocyanins on·OH before and after purification are 96.25% and 97.85% respectively.The scavenging rates of anthocyanins on DPPH·before and after purification are 88.35% and 93.20% respectively.The mixed resins of D101 and HP-20 have good purification effects on blueberry anthocyanins and can be used for the purification of blueberry anthocyanins.After purification,the antioxidant capacity of blueberry anthocyanins is significantly improved,which can better scavenge·OH and DPPH·,and the scavenging effect on·OH is higher than that on DPPH·.
