超高效液相色谱比较不同产地天麻的多酚差异

摘要 通过超高效液相色谱分离检测7个产地天麻的多酚化合物,比较其多酚化合物的质量浓度和差异,辅助评估天麻药材的品质。采用超高效液相色谱-紫外检测和梯度洗脱,多酚化合物在检测中获得良好的分离和峰型;运用此法测定天麻多酚质量浓度,24 h内峰面积稳定,相对标准偏差值1.03%(n=5),较为可靠。陕西、江苏、云南、贵州、四川、广西和吉林7个产地天麻中的天麻素、香草醛、对羟基苯甲酸及丁香醛质量浓度分别为0.341,0.018,0.013,0.026 mg·g^(-1);0.183,0.019,0,0.026 mg·g^(-1);0.207,0.009,0,0 mg·g^(-1);0.238,0.026,0,0.025 mg·g^(-1);0.190,0.018,0,0.013 mg·g^(-1);0.365,0.028,0.026,0.026 mg·g^(-1);0.357,0.028,0.013,0.027 mg·g^(-1)。各地区样品中,均未测到香豆酸、阿魏酸和咖啡酸。天麻的多酚总量由高到低依次为广西>吉林>陕西>贵州>江苏>四川>云南,每种多酚的平均质量浓度由高到低依次为天麻素>香草醛>丁香醛>对羟基苯甲酸,为天麻多酚物质的开发和天麻品质的评估提供一定的理论依据和参考价值。 The aim of this study was to analyze the content of above polyphenols in Gastrodia in different ecological areas by ultra-high pressure liquid chromatography,and to compare the differences of polyphenols in Gastrodia in different areas by analyzing the polyphenol fingerprints,and to evaluate the quality of Gastrodia.The super high-performance liquid chromatography of polyphenols Gastrodia elBlume was determined by ultra-violet detection and gradient elution,and the peak area was stable within 24 hours,the relative standard deviation was 1.03%(n=5).The concentrations of gastrodin,vanillin,p-hydroxybenzoic acid and syringaldehyde in Gastrodia elata from Shanxi,Jiangsu,Yunnan,Guizhou,Sichuan,Guangxi and Jilin were 0.341,0.018,0.013,0.026 mg·g^(-1),0.183,0.019,0,0.026 mg·g^(-1),0.207,0.009,0,0 mg·g^(-1),0.238,0.026,0,0.025 mg·g^(-1),0.190,0.018,0,0.013 mg·g^(-1),0.365,0.028,0.026,0.026 mg·g^(-1),0.357,0.028,0.013,0.027 mg·g^(-1).Coumaric acid,ferulic acid and caffeic acid were not detected in all samples.The sequence of total polyphenols in Gastrodia elata from high to low is Guangxi,Jilin,Shanxi,Guizhou,Jiangsu,Sichuan,and Yunnan.The content of each polyphenol was in the order of gastrodin,vanillin,syringaldehyde and p-hydroxybenzoic acid,which provided some theoretical basis and reference value for the development of Gastrodia polyphenols and the quality identification of Gastrodia.
