红瑞木烂皮病病原鉴定及生物学特性

摘要 2022年在东北林业大学校园内发现了一种红瑞木枝干表皮新病害,主要表现为感病枝条产生水渍斑,直至枯黄和脱落。通过单孢分离法获得病原菌后观察比对病原菌形态特征,并利用核糖体DNA基因(ITS)、核糖体大亚基(LSU)和转录延伸因子(Tef1α)3个基因区域构建多基因系统发育树确定病原物,此外还设置了不同培养基、碳氮源、pH值、温度和光照条件对病原菌的生物学特性做出了初步探究。结果表明:引起该病害的病原菌为Cytospora elaeagni,该真菌为弱寄生菌;最佳培养基为PSA,最适于葡萄糖为碳源、蛋白胨为氮源、25℃、pH为7的全黑暗条件下生长,但在24 h光照和12 h光照与12 h黑暗交替条件下可产生更多分生孢子器。 In 2022,a new disease affecting the twig bark of Cornus alba was discovered on the campus of Northeast Forestry University.The symptoms primarily manifest as water-soaked spots on infected branches,progressing to yellowing and shedding.The pathogen was isolated using single spore isolation method,and its morphological characteristics were observed and compared.Utilizing three genetic regions,namely ITS,LSU of ribosomal DNA,and Tef1α,was employed to construct a multi-gene phylogenetic tree to identify the pathogen.Furthermore,the biological characteristics of the pathogen were preliminarily investigated on different culture media,carbon and nitrogen sources,pH,temperatures,and light conditions.The results showed that the causative agent of this disease is Cytospora elaeagni,a weak parasitic fungus.Optimal growth conditions were found to be on Potato Dextrose Agar(PSA)medium,with glucose as the preferred carbon source,peptone as the nitrogen source,at 25℃under dark conditions(pH=7).However,more conidiomata were produced under 24-hour and 12-hour of light and 12-hour of dark alternations.
