碳酸盐碱度对中华绒螯蟹仔蟹耐受性和 抗氧化物酶活性的影响

摘要 为研究不同碳酸盐碱度对中华绒螯蟹(Eriocheir sinensis)仔蟹成活率和抗氧化酶活性的影响,在不同碱度条件下(7.00、9.49、12.86、17.42、23.61、32.00 mmol/L)对中华绒螯蟹新品种“江海21号”仔蟹进行96 h急性碱度胁迫实验,探讨仔蟹对碳酸盐碱度的适应性。结果显示:中华绒螯蟹仔蟹随着碱度的升高,死亡率增加,在24、48、96 h时的碳酸氢钠碱度半致死浓度LC 50分别为62.32、46.77、34.22 mmol/L,安全浓度SC为7.90 mmol/L;对照组7.00 mmol/L超氧化物歧化酶活性无显著差异,9.49、12.86、17.42、23.61 mmol/L实验组SOD活性均呈现先显著升高后降低的趋势,在24 h达到峰值,而32.00 mmol/L呈现显著增加的趋势;各实验组过氧化氢酶(CAT)活性显著低于对照组,9.49、12.86、17.42 mmol/L浓度碱度组CAT活性随胁迫时间的变化呈现下降趋势。23.61、32.00 mmol/L高浓度碱度组总体呈先降低后升高的趋势;各实验组组织液总抗氧化能力(T-AOC)活性均呈现先显著升高再降低的趋势;32.00 mmol/L浓度组谷胱甘肽过氧化物酶(GSH-Px)活性呈现显著增加的趋势,其它实验组均出现先显著升高再降低的趋势并在24 h时达到峰值。不同碱度对中华绒螯蟹仔蟹生存有较大的影响,中华绒螯仔蟹可以通过激活抗氧化系统来适应一定程度的碱度胁迫。 The area of saline-alkali water in China is about 46000 million hectares.The cultivation in saline-alkali water is one of the ten models of comprehensive utilization of saline-alkali resources.The development of saline-alkaline aquaculture can effectively utilize resources and improve economic and ecological benefits.According to the types and characteristics of saline-alkaline water resources,most of the inland areas in China are carbonate-type waters,especially in Inner Mongolia,Ningxia,Xinjiang,etc.,it is of great significance to explore the best economic varieties suitable for local breeding.Since Eriocheir sinensis is highly adaptable to the environment,can live in saline-alkaline water,and has high economic value.It is essential to study the adaptability of E.sinensis to the alkalinity of carbonate.Alkalinity is an important water quality indicator that affects the growth of E.sinensis,especially for the breeding and promotion of new varieties in saline alkali water areas,which have great significance.The effects of various levels of alkalinity stress(7,9.49,12.86,17.42,23.61 and 32.00 mmol/L)in E.sinensis were investigated utilizing measuring the survival rate and antioxidant enzyme activity of juvenile crabs on the new variety of E.sinensis,"Jianghai 21"at 0,24,48 and 96 h.The results showed that the mortality rate increased as the alkalinity of Chinese mitten crab larvae increased.The median lethal concentration(LC 50)for alkalinity at 24,48,and 96 hours were 62.32,46.77,and 34.22 mmol/L,respectively,and the safe concentration was 7.90 mmol/L.There was no significant difference in the superoxide dismutase(SOD)activity of the control group at 7 mmol/L.In contrast,the experimental groups at 9.49,12.86,17.42,and 23.61 mmol/L showed a significant increase followed by a decrease,reaching its peak at 24 hours,while the 32 mmol/L showed a significant increase.The activities of catalase(CAT)in each experimental group were significantly lower than that of the control group,and the CAT activity of the alkalinity groups at concentrations of 9.49,12.86,and 17.42 mmol/L showed a decreasing trend with the change of stress time.The high-concentration alkalinity groups of 23.61 and 32 mmol/L showed an overall trend of first decreasing and then increasing.The total antioxidant capacity(T-AOC)activities in each concentration group generally showed a trend of a rst increasing and then decreasing during the 96-h stress process,while no signi cant changes were observed in the control group during this period.The activities of glutathione peroxidase(GSH-Px)in the 32 mmol/L concentration group showed a significant increase trend.In contrast,other experimental groups showed a trend of first significantly increasing and then decreasing,reaching a peak at 24 hours.Different alkalinity has a significant impact on the survival of E.sinensis.E.sinensis can adapt to a certain degree of alkalinity stress by activating its antioxidant system.
