早期子宫内膜癌组织及癌旁组织差异蛋白质组学研究

Exploring the Differential Proteomic Analysis of Early Endometrial Carcinoma Tissue and Tumor-adjacent Tissue

摘要 目的筛查Ⅰ期子宫内膜癌及癌旁组织的差异蛋白质,探索其相关的生物标志物,以期用于子宫内膜癌的早期筛查及诊断。方法收集2021年9月至2022年9月于内蒙古自治区人民医院妇科确诊为Ⅰ期子宫内膜癌患者的癌组织及配对癌旁组织标本10对,运用蛋白质谱分析技术,寻找子宫内膜癌与癌旁组织的差异表达蛋白质,并进行生物信息学分析,挑选其中可能成为肿瘤标志物的蛋白质因子。结果在子宫内膜癌组织和远端癌旁组织中共找到了差异表达的蛋白质62个,其中表达显著上调的有37个,表达下调的有25个,其中MGAT4A和FBXO44两个蛋白质在子宫内膜癌组织中呈显著表达上调。结论子宫内膜癌组织与癌旁组织存在差异表达蛋白质,其中MGAT4A和FBXO44蛋白质较正常内膜组织中表达明显升高,有可能是潜在的肿瘤标志物。 Objective:To screen the differential proteins of stage I endometrial carcinoma and its adjacent tissues, and to explore its related biomarkers for the early screening and diagnosis of endometrial carcinoma.Methods Ten pairs of cancer tissue and paired para-cancer tissue samples from patients diagnosed with stage I endometrial cancer were collected from the gynecology department of Inner Mongolia Autonomous Region People's Hospital from September 2021 to September 2022. Protein profiling technology was used to find the differentially expressed proteins in endometrial cancer and para-cancer tissues, and bioinformatics analysis was performed.Select the protein factors that may be tumor markers.Results A total of 62 proteins were differentially expressed in endometrial carcinoma tissues compared with adjacent tissues, 37 proteins were up-regulated and 25 proteins were down-regulated.The Alpha 1,3-mannosyl-glycoprotein 4-beta-N-acetylglucosaminyltransferase A, MGAT4A and F-box protein 44,The expression of FBXO44 was significantly up-regulated in endometrial carcinoma.Conclusion There are different expression proteins in endometrial carcinoma tissues and adjacent tissues The expression of MGAT4A and FBXO44 proteins in endometrial carcinoma tissues was significantly higher than that in normal endometrial tissues, which may be potential tumor markers.
