摘要
胶原蛋白是一种生物功能蛋白,广泛应用于生物医学、化妆品护肤、食品、新型材料等诸多领域。重组胶原蛋白因其避免了免疫反应,且产量和性能稳定,具有替代动物提取胶原蛋白的巨大潜力。然而,重组胶原蛋白在工业应用中仍面临着表达量低、工业生产成本高和羟化难等挑战。重点阐述了重组胶原蛋白发酵过程中影响产量的关键因素,结合国内外的实验研究,得出每个关键因素适宜的控制条件;介绍了重组胶原蛋白纯化过程常用的制备工艺及机理,并结合实例探讨了每种工艺对提取重组胶原蛋白的实际效果;最后对重组胶原蛋白的发酵和纯化工艺的优化进行了展望。
Collagen is a biofunctional protein that has been widely used in many fields,including but not limited to biomedical,cosmetics and skin care,food,and novel materials.Recombinant collagen has great potential as an alternative to collagen extracted from animals because it avoids the immune response,also the yield and properties are stable.However,recombinant collagen still faces challenges in industrial applications such as low expression,high cost in industrial production and difficult hydroxylation.This paper focuses on the key factors affecting the yield of recombinant collagen fermentation,and obtains the appropriate control conditions for each key factor based on the domestic and foreign experimental researches.The common preparation technology and mechanism of recombinant collagen purification process were introduced,and the practical effect of each process on the extraction of recombinant collagen was discussed with examples.Finally,the optimization of fermentation and purification process of recombinant collagen was prospected.
