2019—2023年河南省某市食品中致泻大肠埃希菌污染状况及毒力基因分析

摘要 目的了解河南省某市食品样品中致泻大肠埃希菌(diarrheagnic Escherichia coli,DEC)污染状况及毒力基因分布,为制定食源性疾病防控措施提供依据。方法按照《河南省食品中微生物及其致病因子监测工作手册》,对2019—2023年某市采集的339份食品样本进行分析,对DEC进行分离、鉴定和基因分型,并做毒力基因分型。结果食品样本中DEC总检出率为9.7%(33/339),不同年份的检出率差异有统计学意义(χ^(2)=12.626,P<0.05)。不同食品类别检出率差异有统计学意义(χ^(2)=30.795,P<0.05),其中生鲜肉类食品检出率最高(20.4%);散装食品检出率高于包装食品(χ^(2)=29.364,P<0.05)。分离鉴定出36株DEC,肠聚集性大肠埃希菌是优势毒力株,携带毒力基因以集聚热稳定性毒素A基因和肠定植因子基因为主。结论某市食品中致泻大肠埃希菌污染较严重,肠聚集性大肠埃希菌是食品污染的优势毒力株。 Objective To investigate the prevalence and distribution of virulence genes in diarrheagnic Escherichia coli(DEC)in food samples from a city in Henan Province,so as to provide data to inform the development of foodborne disease prevention and control strategies.Methods Following Guidelines of the Handbook of Monitoring Microbes and Their Pathogenic Factors in Food in Henan Province,a total of 339 food samples collected from 2019 to 2023 were analyzed.DEC strains were isolated,identified,and genotyped the virulence genes.Results The overall detection rate of DEC in food samples was 9.7%(33/339).Statistically significant variations were observed in detection rates across different years(χ^(2)=12.626,P<0.05).The detection rates among different food categories were statistically significant(χ^(2)=30.795,P<0.05)with the highest detection rate in fresh meat samples(20.4%).Bulk food samples had a higher detection rate compared to packaged food(χ^(2)=29.364,P<0.05).Thirty-six virulent DEC strains were isolated and characterized,with enteroaggregative Escherichia coli(AECE)was the dominant strain.The primary virulence genes identified were the aggregative heat-stable toxin A gene and intestinal colonization factor gene.Conclusion Food in the studied city is significantly contaminated with DEC,with AECE is the dominant virulence strain of food contamination.
