基于氧化还原体系纸芯片法快速检测血清葡萄糖含量

摘要 建立了一种以纸芯片为分析平台的葡萄糖含量快速检测新方法。利用葡萄糖的还原性,以过碘酸为氧化剂组成氧化还原体系,以希夫试剂为显色剂对反应产物进行显色,根据显色强度对葡萄糖含量进行测定。结果表明,葡萄糖、过碘酸和希夫试剂最佳用量分别为6,4和6μL,反应5 min时显色最佳。在2~10 mmol/L范围内,显色强度与葡萄糖浓度呈良好线性关系,相关系数r为0.9972。该方法的检测限为4μmol/L,日内和日间相对标准偏差(RSD)分别为1.1%和1.3%。6种共存物质的干扰误差均在-0.1%~2.6%。对115份血清样品的测定结果与医院检测值之间无显著性差异。该方法可用于血清中葡萄糖含量检测。 A novel method for rapid determination of glucose was developed based on a microfluidic paper-based analytical device using a redox system.Herein,glucose reacts with periodic acid to form a product with an aldehyde group,which subsequently reacts with Schiff reagent to form the red complex.The color intensity was recorded to analyze the concentration of glucose.The results showed that the optimal loading volumes for glucose,periodic acid and the Schiff reagent were 6,4 and 6μL,respectively,with an optimum reaction time of 5 min.A good linear relationship was obtained between color intensity and glucose concentration in the range of 2-10 mmol/L,with a correlation coefficient(R)of 0.9972.The limit of detection(LOD)of this method was 4μmol/L,and the relative standard deviations(RSDs)of intra-day and inter-day were 1.1%and 1.3%,respectively.The interference deviations of 6 coexisting substances were in the range of-0.1%to 2.6%.There was no significant difference between the results of 115 serum samples obtained by this method and the hospital test results.This method can be used for the determination of glucose in serum.
