摘要
建立了液相色谱-串联质谱法同时测定猪可食组织中氯丙嗪、异丙嗪、甲苯噻嗪等精神活性物质及其代谢物的方法。通过构建回收率及相对标准偏差的多元质量特性双响应曲面模型进行过程能力分析,对QuEChERS方法进行优化,并结合同位素内标法进行检测。结果表明,在质量浓度0.40~200 ng/mL范围内7种目标化合物呈良好的线性关系,相关系数(r^(2))均大于0.999,方法检出限范围为0.05~0.10μg/kg,定量限范围为0.20~0.40μg/kg。5种不同基质中7种目标化合物的加标回收率在78.5%~109%之间,相对标准偏差(RSD)在0.89%~10.8%之间,10日内方法重现性RSD在2.3%~12%之间。所建立的方法适用于猪可食组织中几种常见精神活性物质及其代谢物的定量分析检测。
A method for the determination of psychoactive substances and their metabolites such as chlorpromazine,promethazine and xylazine in edible tissues of swine was developed by using high-performance liquid chromatographytandem mass spectrometry(HPLC-MS/MS).The pre-treatment method of QuEChERS was optimized by constructing a multivariate mass characteristic double response surface model of recovery and relative standard deviation(RSD)for process capability evaluation,and the isotope internal standard method was used simultaneously for sample detection.The results showed that good linear relationships with the correlation coefficients(r^(2))greater than 0.999 in the mass concentration range of 0.40-200 ng/mL can be achieved for seven target compounds.This method exhibits excellent detection performance with low limit of detection(LODs)of 0.05-0.10μg/kg and the limit of quantification(LOQs)of 0.20-0.40μg/kg.The recoveries of the seven target compounds in five different matrices were 78.5%-109%,with the relative standard deviations(RSDs)of 0.89%-10.8%.Besides,the reproducibility of this assay within 10 d ranged from 2.3%to 12%.The proposed method is suitable for the quantitative analysis of several common psychoactive substances and their metabolites in the edible tissues of swine.
