摘要
为明确河北省部分地区大肠埃希氏菌携带的毒力因子情况及对药物的敏感度,更好地控制由致病性大肠埃希氏菌导致的犊牛腹泻,从河北省部分地区(廊坊、定州、石家庄、保定、衡水、邢台)的牛场采集了128份腹泻犊牛粪样,从中分离大肠埃希氏菌,利用PCR对分离的大肠埃希氏菌进行毒力因子和耐药基因的检测,并采用K-B纸片法对分离菌株进行12种抗菌药物的药物敏感性试验。结果显示,从128份腹泻犊牛粪便中分离到菌株125株,其中104株为大肠埃希氏菌。104株分离菌中61株携带毒力基因,毒力基因携带率为58.70%(61/104),毒力基因F17、Stx1、irp2、FyuA、STa、Stx2阳性率分别为17.31%、0.08%、24.04%、35.58%、10.58%、0.04%,未检测出K99、F41、LT和eaeA基因,部分大肠埃希氏菌携带多重毒力因子。16种耐药基因的检测显示aph(3'')-Ib、tetC、sul3、qnrA、qnrC、tetB、aac(6′)-Ⅰb、tetA、bla TEM、bla CTX-M、sul1、sul2、oqxA、oqxB、qnrS、qnrB阳性率分别为59.62%、26.0%、28.85%、0.05%、32.69%、29.81%、46.15%、48.08%、82.69%、78.85%、55.77%、74.04%、25.00%、30.77%、43.27%、0.07%。药物敏感性试验结果显示,104株分离菌耐2种以上抗菌药物的菌株达86.62%,对卡那霉素、恩诺沙星和环丙沙星敏感率较高,研究结果可为河北省部分地区大肠埃希氏菌性犊牛腹泻的防治提供理论依据。
In order to clarify the situation of virulence factors carried by Escherichia coli and its sensitivity to drugs in some areas of Hebei province,and to better control calf diarrhoea caused by pathogenic E.coli,128 feces of diarrhoeic calves were collected from cattle farms in some areas of Hebei province(Langfang,Dingzhou,Shijiazhuang,Baoding,Hengshui and Xingtai)from which E.coli was isolated,and the isolated E.coli were subjected to the detection of virulence factors and drug-resistance genes by PCR,and drug-sensitivity tests of 14 antimicrobials by KB paper method for the isolated strains.The results showed a total of 125 strains were isolated from the faeces of 128 diarrhoeic calves,of which 104 strains were E.coli,61 of the 104 isolates carried virulence genes,the virulence gene carriage rate was 58.70%(61/104),and the virulence genes F17,Stx1,irp2,FyuA,Sta and Stx2 were positive in 17.31%,0.08%,24.04%,35.58%,10.58%,and 0.04%,respectively,and no K99,F41,LT and eaeA genes were detected,and some E.coli carried multiple virulence factors.The detection of 16 resistance genes showed that positive rates of aph(3″)-Ib,tetC,sul3,qnrA,qnrC,tetB,aac(6′)-Ⅰb,tetA,bla TEM,bla CTX-M,sul1,sul2,oqxA,oqxB,qnrS,qnrB were 59.62%,26.0%,28.85%,0.05%,32.69%,29.81%,46.15%,48.08%,82.69%,78.85%,55.77%,74.04%,25.00%,30.77%,43.27%and 0.07%,respectively.The results of drug sensitivity test showed that 104 isolates were resistant to more than 2 antimicrobial drugs up to 86.62%,and the sensitive rate to kanamycin,enrofloxacin and ciprofloxacin was higher.This study can provide theoretical basis for the prevention and control of E.coli calf diarrhoea in some areas of Hebei province.
