摘要
D-塔格糖(D-tagatose)是一种稀有的天然六碳酮糖,是D-半乳糖同分异构体、D-果糖C-4位置上的差向异构体,学术界把这类天然存在较为罕见的单糖称为稀有糖。本文简要介绍了生物催化法合成D-塔格糖的研究背景和意义、D-塔格糖的物理化学性质和生理功能,并重点介绍了生物合成D-塔格糖主要的生物酶L-阿拉伯糖异构酶(L-arabinose isomerase,L-AI)的性质及结构和L-阿拉伯糖异构酶N-糖基化分子改良,并对未来D-塔格糖的生物合成研究做出展望,以期为D-塔格糖进行工业产业化提供参考价值。
D-tagatose is a rare natural six carbon ketose.It is a D-galactose isomer and an epimer of D-fructose at the C-4 position.This type of rare monosaccharide with limited natural occurrence is known as rare sugar in academia.This paper briefly introduced the research background and significance of biocatalytic synthesis of D-tagatose,the physicochemical properties and physiological functions of D-tagatose,and focused on the properties and structure of main biocatalyst L-arabinose isomerase(L-AI),and the improvement of L-arabinose isomerase N-glycosylation molecule.The future research direction of biocatalytic synthesis of D-tagatose was also discussed to provide reference value for the industrialization of D-tagatose.
