摘要
铱(Ⅲ)抗肿瘤配合物[(η^(5)-C_(5)MeC_(6)H_(4)C_(6)H_(5))Ir(C∧C)Cl]PF_(6)能够选择性杀伤、抑制肿瘤细胞,但其内在的作用机制尚不完全明确.采用细胞划痕法、迁移及侵袭等实验考察了金属铱(Ⅲ)配合物对A549细胞的抗转移能力,采用免疫组织化学法和蛋白印迹法分析NEDD9、β-catenin表达水平.研究结果显示:金属铱(Ⅲ)配合物对A549细胞有明显的抗转移作用,可能通过作用于NEDD9降低肺癌细胞的增殖和转移.同时该金属配合物也可通过诱发β-catenin蛋白的表达,增加细胞膜表面粘性,降低A549细胞的转移能力.
The designed and developed iridium(Ⅲ)anticancer complex[(η^(5)-C_(5)MeC_(6)H_(4)C_(6)H_(5))Ir(C∧C)Cl]PF_(6)can selectively kill and inhibit tumor cell,but its internal mechanism of action is not fully understood.The anti-metastasis ability of iridium(Ⅲ)complex on A549 cells was investigated by cell scratch,migration and invasion assay.The expression levels of NEDD9 andβ-catenin were analyzed by immunohistochemistry and western blot.The results showed that the iridium(Ⅲ)complex had a significant anti-metastasis effect on A549 cells,which may reduce the proliferation and metastasis of lung cancer cells by acting on NEDD9.At the same time,the gold complex can also induce the expression ofβ-catenin protein,increase the membrane surface viscosity,and reduce the metastasis ability of A549 cells.In this study,cell scratch method,migration and invasion experiments were used to investigate the anti-metastasis ability of the metal iridium(Ⅲ)complex to A549 cells.The expression levels of NEDD9 andβ-catenin were analyzed by immunohistochemistry and western blot.The results show that the metal iridium(Ⅲ)complex has obvious anti-metastasis ability to A549 cells and may reduce the proliferation and metastasis of lung cancer cells by acting on NEDD9.And the complex has also induced the expression ofβ-catenin protein,increased the membrane surface viscosity,and reduced the transfer capacity of A549 cells.
