基于生物信息学方法设计鼠伤寒沙门氏菌OmpC-FliC多表位疫苗

Design of Salmonella typhimurium OmpC-FliC multi-epitope vaccine based on bioinformatics method

摘要 为了设计鼠伤寒沙门氏菌OmpC-FliC多表位疫苗,试验在NCBI网站的Protein数据库中获取OmpC和FliC蛋白的氨基酸序列,采用Expasy服务器分析其理化性质,通过在线工具AllerTopV.2.0和Vexijen分析其抗原性和致敏性,应用NPS~@服务器分析二级结构,应用IEDB在线工具分析OmpC和FliC的B、T细胞表位。选取优势表位设计鼠伤寒沙门氏菌OmpC-FliC多表位疫苗,对其理化性质、抗原性、致敏性及三级结构进行预测。结果表明:OmpC和FliC均是亲水性蛋白,无致敏性,抗原性较高;OmpC和FliC蛋白均筛选出3个B细胞优势表位,2个细胞毒性T细胞(CTL细胞)优势表位,1个辅助T细胞(Th细胞)优势表位。设计的多表位疫苗为亲水性的稳定蛋白,抗原性预测的分数为1.1422分,且为非致敏原,预测三级结构模型的可信度大。说明鼠伤寒沙门氏菌OmpC-FliC多表位疫苗对动物无致敏性,且具有良好的抗原性。 In order to design a Salmonella typhimurium OmpC-FliC multi-epitope vaccine,in the experiment,the amino acid sequences of OmpC and FliC proteins were obtained from the Protein database on the NCBI website;the physicochemical properties were analyzed using the Expasy server;the antigenicity and sensitization were analyzed by the online tool AllerTopV.2.0 and Vexijen;the secondary structure was analyzed by applying the NPS~@server;and the IEDB online tool was applied to analyze and B and T cell epitopes of OmpC and FliC.The dominant epitopes were selected to design Salmonella typhimurium OmpC-FliC multi-epitope vaccine,and their physicochemical properties,antigenicity,sensitization and tertiary structure were predicted.The results showed that both OmpC and FliC were nonallergenic hydrophilic proteins with high antigenicity;both OmpC and FliC proteins were screened for three B-cell-dominant epitopes,two cytotoxic T-cell(CTL cell)dominant epitopes and one helper T-cell(Th cell)dominant epitope.The designed multi-epitope vaccine was a hydrophilic,stable protein,with an antigenicity prediction score of 1.1422,and was a non-sensitizer;the predicted tertiary structural model had high confidence.The results suggested that the Salmonella typhimurium OmpC-FliC multi-epitope vaccine was non-allergenic and had good antigenicity in animals.
