摘要
为培育铁线莲新品种、丰富其栽培类型,以大叶铁线莲、卷萼铁线莲、短尾铁线莲3种不同生长类型的野生铁线莲为亲本进行远缘杂交育种并鉴定其子代的真实性,探究子代与亲本的遗传变异规律和遗传多样性。结果表明:利用12条ISSR引物对3个亲本的6个杂交组合的子代进行真实性鉴定,47个子代鉴定出了43个真杂种。杂交亲本及子代的等位基因数变化范围为1.731~1.937,有效等位基因数变化范围为1.449~1.613,期望杂合度变化范围为0.273~0.352,多样性指数在0.417~0.527之间,表明子代具有丰富的遗传变异。杂交子代部分表型性状受到了亲本的影响,在直立和藤蔓2种亲本生长类型的基础上出现了匍匐生长类型的子代,萼片长度、萼片宽度、花径、叶片宽度呈现增加趋势,其余花部和叶部性状则相反。表型性状结合分子标记可有效用于野生铁线莲杂交子代早期真实性鉴定,缩短育种周期,为新品种选育提供理论依据。
In order to cultivate new Clematis varieties and enrich its cultivation types,three wild Clematis species were used as parents for cross-breeding.We identified the true hybrids of their progenies,and explored the genetic variation patterns and genetic diversity between the progenies and the parents.The results showed that 43 true hybrids were identified in 47 progenies from six hybrid combinations using 12 ISSR primers.The allele number of parents and progenies varied from 1.731 to 1.937,the effective allele number varied from 1.449 to 1.613,the expected heterozygosity varied from 0.273 to 0.352,and the diversity index ranged from 0.417 to 0.527,which indicated that the progenies were rich in genetic variation.Some of the phenotypic characters in the hybrid progenies were influenced by the parents with variable growth types,including an increasing trend in sepal length,sepal width,flower diameter,and leaf blade width,while the rest of the floral and foliar traits were opposite.The combination of phenotypic character and molecular markers can be effectively used for the early authenticity identification of wild Clematis hybrid offspring,shorten the breeding cycle,and provide a theoretical foundation for the selection of new varieties.
