摘要
目的建立RP-HPLC-一测多评法(QAMS)同时测定左金丸中不同类型生物碱(盐酸小檗碱、吴茱萸碱和吴茱萸次碱)的含量。方法采用70%乙醇作提取剂对样品进行加热回流提取,色谱柱为MG C_(18)(4.6 mm×250 mm,5μm);流动相是乙腈(A)-0.1%磷酸水溶液(B);流速:1.0 mL·min^(-1);柱温35℃;检测波长有3个,分别为264 nm(盐酸小檗碱)、226 nm(吴茱萸碱)、342 nm(吴茱萸次碱)。结果盐酸小檗碱的进样量在8.68~173.521μg、吴茱萸碱的进样量在1.04~20.83μg、吴茱萸次碱的进样量在1.06~21.19μg,与峰面积有良好的线性关系(r≥0.999),加样回收率依次是99.0%、98.8%、100.4%。吴茱萸碱、吴茱萸次碱与盐酸小檗碱之间的相对校正因子分别为2.053和1.671。一测多评法其测出的结果与外标法接近。结论该方法计算结果准确,可靠,可用于左金丸中3种生物碱的含量测定。
OBJECTIVE A method was developed to determine the quality of the different types of alkaloids(berberine hydrochloride,evodiamine and rutaecarpin)by RP-HPLC and quantitative analysis of multi-components with single maker(QAMS)in ZuoJin Pills.METHODS The samples were extraced with 70%ethanol and heated to reflux in water bath.The MG C_(18)column(4.6 mm×250 mm,5μm)was used.The mobile phase was consisted of acetonitrile(A)-aqueous solution containing 0.1%phosphoric acid(B).The flow rate of the mobile phase was 1.0 mL·min^(-1).The column temperature was set at 35℃.The alkaloids were monitored at three different detection wavelengths which 264 nm for berberine hydrochloride,226 nm for evodiamine and 342 nm for rutaecarpin.RESULTS The injection volume of alkaloids displayed well linear range within the range of 8.68-173.521μg for berberine hydrochloride,1.04-20.83μg for evodiamine and 1.06-21.19μg for rutaecarpin.The recoveries of three alkaloids,berberine hydrochloride,evodiamine and rutaecarpin,were 99.0%,98.8%,100.4%,respectively.The relative correction factor between evodiamine and berberine hydrochloride,rutaecarpin and berberine hydrochloride were 2.053 and 1.671,respectively.The results were close to that of the external standard method.CONCLUSION The method is accurate and reliable,it can be used for simultaneous determination of three alkaloids in ZuoJin Pills.
