免疫亲和柱净化-高效液相色谱-串联质谱法测定大豆油中的4种黄曲霉毒素

Determination of Four Aflatoxins in Soybean Oil by High Performance Liquid Chromatography-Tandem Mass Spectrometry with Immunoaffinity Column Cleanup

摘要 本文采用免疫亲和柱净化-高效液相色谱-串联质谱法建立了一种测定大豆油中4种黄曲霉毒素(B_(1)、B_(2)、G_(1)和G_(2))的分析方法。试样经免疫亲和柱分离,再经高效液相-串联质谱测定,通过外标法定量,进行精密度和回收率的实验验证。结果表明,4种黄曲霉毒素在各自浓度范围内线性关系良好,相关系数(R^(2))均大于0.9950,检测限为0.070~0.021μg·kg^(-1),定量限为0.06~0.20μg·kg^(-1)。加标回收率为88.7%~108.5%,相对标准偏差为0.3%~7.2%。该方法净化效率好,准确度高,灵敏度高和耐受性好,可用于大豆油中4种黄曲霉毒素的准确检测。 This article establishes an analytical method for the determination of four aflatoxins(B_(),B_(2),G_(1),and G_(2))in soybean oil using immunoaffinity column purification high performance liquid chromatography tandem mass spectrometry.The sample was separated by an immunoaffinity column and then determined by high-performance liquid chromatography tandem mass spectrometry.Quantification was performed using an external standard method to experimentally verify the precision and recovery rate.The results showed that the four types of aflatoxins had good linear relationships within their respective concentration ranges,with correlation coefficients(R^(2))greater than 0.9950.The detection limit was 0.070~0.021μg·kg^(-1),and the quantification limit was 0.06~0.20μg·kg^(-1).The recovery rate of spiked samples ranges from 88.7%to 108.5%,with a relative standard deviation of 0.3%to 7.2%.This method has good purification efficiency,high accuracy,high sensitivity,and good tolerance,and can be used for accurate detection of four types of aflatoxins in soybean oil.
